objectives :To detect the expression level of Transglutaminase 2(TG2)and Human papilloma virus 16(HPV16)-E6 in normal cervical tissues, cervical intraepithelial neoplasias(CIN)tissues and cervical squamous cell carcinoma(CSCC) tissues. To investigate the relationship between expression of TG2 and HPV16-E6 and the correlation among expression of TG2 and HPV16-E6 and the clinic pathological characters in CSCC.Methods 1. The expression level of TG2 and HPV16-E6 were detected by Western Blot, the m RNA level were detected by RT-PCR, in 40 cases of normal cervical tissues, 48 cases of CIN and 48 cases of CSCC.2. TG2 and HPV16-E6 expression were measured by immunohistochemical stain(IHC) in 40 cases of normal cervical tissues, 48 cases of CIN and 48 cases of CSCC.3. The relationship between HPV16-E6 and TG2 was analysized by Spearman rank.4. The relationship among TG2 and HPV16-E6 and the prognosis of CSCC were analyzed by Kaplan-Meier method.Results: 1. Compare to normal cervical tissues, the expression of TG2, HPV E16 and its m RNA were all higher in CSCC. There were significant difference between normal cervical tissues and CSCC(P<0.05). And there were no significant difference in the expression of TG2, HPV E16 and its m RNA between normal cervical tissues and CSCC(P>0.05).2. In CSCC, the expressions level of TG2 were correlated with the clinical stage, lymph node metastases, infiltrating depth(P<0.05), but without relationship to age, histologic grade(P>0.05). In CSCC, the expression level of HPV16 were correlated with age(P<0.05), but without relationship to the histological grade, clinical stage, degree of differentiation, lymph node metastasis and invasion depth(P>0.05).3. The expression of TG2 and HPV16-E6 in normal cervical tissues, CIN and CSCC increased gradually. There were significant difference in the positive rate of TG2 and HPV16-E6 among normal cervical tissues and CIN III and CSCC(P<0.05), and without significant difference among normal cervical tissues and CIN I and II(P>0.05). There were significant difference in the positive rate of TG2 and HPV16-E6 among CIN I,II and CSCC(P<0.05), and without significant difference among normal cervical tissues and CIN I, II and III(P>0.05).4. In CSCC, TG2 was positively associated with HPV16-E6 expression(r=0.405, P=0.006).5. The survival rate were lower in the patients with TG2 positive than TG2 negative. There were significant difference between them(P=0.009). The survival rate were lower in the patients with HPV16-E6 positive than HPV16-E6 negative. There were no significant difference between them(P>0.05).Conclusions: 1.The expression of TG2 and HPV16-E6 in normal cervical tissue, CIN tissues and CSCC increased gradually. It mean that TG2 and HPV16-E6 had relationship with development and progress of CSCC.2. The expression of TG2 in CSCC with clinical stage, lymph node metastasis and depth of invasion. TG2 can be served as a biomarker for clinic diagnosis and can be served as a biomarker for prognosis in CSCC.
