Objective: Study on intervention effects of Calcium, magnesium, zinc, copper, vitamin D and protein on the apoptosis of ovarian granular cells caused by coal-burning fluorosis in female rats. Methods: 1.A total of 120 female rats were randomly divided into four groups, to establish models in female rats by feeding them corn powder baked by high fluoride coal with mixing different formulations Calcium, magnesium, zinc, copper, vitamin D and protein;2. To observe dynamically the change of female rats,teeth;3.The fluoride contents of urine and bone were measured by ion selective electrode method;4. To dissect the estrum rats and take out ovarian in in different period of time(60d, 120 d, 180d),and the level of apoptosis in ovarian granular cells were detected with TUNEL assay, the expression of Fas and Caspase-3 of in ovarian granular cells were detected with immunohistochemistry,the ultrastructure of ovarian granular cells were observed by transmission electron microscopy,the contents of SOD and MDA were detected by colorimetric.Results: 1.The animal model of fluorosis and intervention were successfully established;2.In the same time group, the results of SOD was the control group > the intervention group with multidimensional element and protein > the intervention group with multidimensional element > the fluorosis group(P<0.05),and the results of MDA 、Fas、Caspase-3 and the apoptosis rate were the fluorosis group>the intervention group with multidimensional element> the intervention group with multidimensional element and protein >the control group; The expression of Caspase-3 reduced gradually with the exposure of time except the intervention group with multidimensional element and protein in 120 d and 180d(P<0.05).Conclusion: 1.Fluorosis can cause the apoptosis of granular cells,and one ofthe mechanisms may be that oxidative stress enhanced the expression of Fas and Caspase-3;2. The Calcium、magnesium、copper、zinc and vitamin D can protect against the fluorosis and prevent the apoptosis of ovarian granular cells by increasing the activity of SOD,decreasing the content of MDA、Fas 、Caspase-3 and the fluoride contents of body,reducing the injury on structure of ovarian granular cells,and the intervention effect was better combined with protein;3. The antagonism has no significant correlation with the exposure of time.
