Purpose: Screening periwinkle(Catharanthus roseus(l.) G.D on) the grass has hypoglycemic active site, and the separation, purification and identification of the active site.Establish periwinkle in total triterpenes and flavonoids content of ultraviolet-visible spectrophotometer determination method, for the overall quality of the periwinkle medicinal material control to provide reliable method.Method: By db/db male mice, the periwinkle grass all petroleum ether extract parts and ethyl acetate extract parts, 95% ethanol extract parts, 70% ethanol extract and 70% ethanol extract of anti-diabetic activity screening.Use of organic solvents extraction and macroporous resin column chromatography method to extract and periwinkle grass all preliminary separation;Using open positive phase silica gel column chromatography, polydextran gel column chromatography and recrystallization method to periwinkle whole grass anti-diabetic, isolation and purification of the chemical composition in the active site by physical and chemical methods and the modern spectrum technology(1 h- NMR, 13 c NMR, ESI- MS) identification of the monomer compounds structure;The uv- spectrophotometric method for periwinkle determination of total triterpenes and flavonoids content.Result: From changchun flower all grass petroleum ether extract and ethyl acetate extract and 95% ethanol extraction part, 70% ethanol extract and 30% ethanol extracted parts, petroleum ether extract parts selection, ethyl acetate extract parts and 95% ethanol extraction parts with anti-diabetic activity, and ethyl acetate extracted parts effect best.And the ethyl acetate part six purified compounds, among them, compound VI by spectrum analysis, determine the steroid and fatty acid mixture.The remaining five compounds by the spectral identification respectively, among them, a long chain fatty acids, 2 triterpene, two sterols, respectively is: 28- alkoxide(octacosanol- 1, Ⅰ);Alpha incense resin alcohol(alpha amyrin Ⅱ);Beta sitosterol(beta sitosterol, Ⅲ);Stigmasterol(stigmasterol, Ⅳ);Ursolic acid, ursolic acid, Ⅴ).Through influence factor and relevant methodology to investigate, determine the periwinkle across the grass, the determination method of total triterpenes and flavonoids content in the method accuracy, precision, stability, good reproducibility, and 5 batch periwinkle medicinal materials by the method for determination of total triterpenes and flavonoids content, total triterpenoid content in ursolic acid was 2.494 ~ 3.953%;Total content of flavonoids in rutin was 0.084 ~ 0.097%.Conclusion: Periwinkle petroleum ether extract parts of the whole grass and ethyl acetate extraction, 95% ethanol extract parts, 70% ethanol extract and 30% ethanol extraction area is significant fall blood sugar function in ethyl acetate parts extracted parts, and six compounds were isolated, according to the physical and chemical properties and spectral data of five compounds were identified, among them, a long chain fatty acids, 2 triterpene, two sterols, respectively is: 28- alkoxide(octacosanol- 1, Ⅰ);Alpha incense resin alcohol(alpha amyrin Ⅱ);Beta sitosterol(beta sitosterol, Ⅲ);Stigmasterol(stigmasterol, Ⅳ);Ursolic acid, ursolic acid, Ⅴ).
