Experimental Study On Fufangzhenhuaxiaoshen’an Capsule’s Effect To Experimental Diabetic Rats’ Podocyte’s Podocin Expression

Purpose: The experiment on the basis of The specialized research fund for the doctoral program of higher education-- From the adiponectin JieDuTongLuoBaoShen Capsule’s Intervention inflammation mechanism of diabetic nephropathy,through observation Fufangzhenhuaxiaoshen Capsule’s intervention effect on diabetic nephropathy rats and influence on glomerular podocytes Podocin expression, which is exploring a new way of prevention and cure of diabetic nephropathy.Method: Using STZ(50mg/Kg) diabetic rats model was established. Diabetic rats were divided into model group,Irbesartan Tablets group(20mg/kg?d),Fuzhenhuaxiaoshen Capsule low dose group(0.25g/Kg · d), Fuzhenhuaxiaoshen Capsule dose group(0.5g/Kg · d),Fuzhenhuaxiaoshen Capsule high dose group(1g/Kg ·d), another 10 normal Wistar rats as normal group, continuous lavage for 12 weeks, normal group and DM model group to capacity such as distilled water to fill the stomach to daily medicine. Detection of each weight of rats、kidney index、urine output、the urine trace albumin levels, determination of blood glucose、Scr、Bun、TC、TG. By HE staining and transmission electron microscope each rat kidney pathological changes, by using immunohistochemistry and rt-pcr detection of Podocin expression in kidney tissue.Result: 12 weeks after treatment, compared with normal group,model group rats’ kidney index、urine output、the urine trace albumin levels、blood glucose、Scr、Bun、TC、TG significantly increased(P<0.01), weight 、 kidney Podocin expression is decreased obviously(P<0.01). Irbesartan Tablets group、Fufangzhenhuaxiaoshen Capsule High, medium and low dose group of rat kidney index、urine output、the urine trace albumin levels、blood glucose、Scr、Bun、TC、TG are all Lower than the model group(P<0.01), weight、kidney Podocin expression are all Higher than that of model group(P<0.01), renal pathological changes obviously improved.Conclusion: Fufangzhenhuaxiaoshen Capsule for early diabetic nephropathy rats kidney plays a protective role, the mechanism may be associated with increased induced Podocin expression.
