Purpose: Detection and analysis of the expression and distribution of the AchR i n EAMG rat neuromuscular junction are conducted by the application of CLSM detect ion technology to explore therapeutic effects and mechanism of acupuncture treatment of "warming yang and invigorating qi" with MG, which provide support and experimenta l basis for acupuncture treatment of MG in clinical practice.Method: 70 SPF grade female Lewis rats were selected, in which 10 Lewis rats were randomly selected as blank group and other 60 SPF grade female Lewis rats were used to establish animal model of EAMG rats with AChR Alpha 1 129-145 more peptide fragment immune vaccination. 30 successful modeling EAMG rats were randomly selected, which divided into model control group of 10 rats, drug control group of 10 rats, and acupuncture treatment group of 10 rats. Blank group was raised in the same period under the same conditions without any processing. Model control group was just clean feeding and did not give any interventions after modeling. Drug control group was treated by intragastric administration with pyridostigmine bromide. Acupuncture treatment group was treated with “warming yang and invigorating qi” acupuncture treatment. After the treatment, fluorescent immunohistochemistry was conducted, and confocal laser scanning microscope(CLSM) was used to collect figures. Image—Pro Plus 6 image analysis software was used to do the analysis and determined the average value of area and accumulative fluorescence density value of the AchR distribution at neuromuscular junction in viewed EAMG rats.Results:(1) 32 animal model of EAMG rats were successfully established. Success rate of establishing model was 53.3%. AchR expression and distribution level of model control group EAMG rats at neuromuscular junction decreased significantly compared with the blank control group with statistical significance(P<0.01);(2) After treatment, AchR expression and distribution expression level of acupuncture treatment group EAMG rats at neuromuscular junction increased obviously compared with model control group with statistical significance(P<0.01);(3) The comparison of acupuncture treatment group and drug control group showed that AchR expression and distribution level of two kinds of treatment methods for EAMG rats at neuromuscular junction resulted similar with no significant difference(P > 0.05).Conclusion:(1) “Warming yang and invigorating qi” acupuncture treatment of EAMG had good therapeutic effect, which was similar to the effect of pyridostigmine bromide.(2) “Warming yang and invigorating qi” acupuncture treatment of MG has good therapeutic effect. The therapy is likely to play a role in treating MG by means of increasing the expression level of AchR at neuromuscular junction.
