Aims This study is explorer the effects of mi R-185 and its mechanism on proliferation and apoptosis in gastric cancer cell lines MGC803 by Targeting CDC42.Methods According to the analysis of related mi RNA by previous result of mi RNA microarray and biological information software, different mi RNA expressed between gastric carcinoma and normal gastric mucosal epithelial cells and possible target gene were selected. Meanwhile the results of mi RNA microarray can be certificated by Real-time PCR. Make mi R-185 mimic and Sh NC plasmid immediately transfected to MGC-803 respectively. Reversed Fluorescence microscope and real-time PCR tested the efficiency of transfection. CCK-8 explored activity of cells and drawed the cell growth curve; cell colony formation explored cell proliferation; Flow Cytometry explored cell cycle and cell apoptosis; Real-time PCR and Western blot explored m RNA of CDC42 and the level of protein expression.Results Mi R-185 selected as the research target by previous result of mi RNA microarray and biological information, and CDC42 probably as target gene, real-time PCR testified that the number of mi R-185 in Gastric cancer cell lines MGC803, SGC7901 decreased, compared with that in normal Gastric mucosal epithelial cells. The result of reversed fluorescence microscope and real-time PCR showed that, in immediately transfected mi R-185 mimic of MGC803 cell, the level of mi R-185 expression increased dramatically. The result of CCK-8 test demonstrated that mi R-185-mimic group restrained cell proliferation obviously. The result of cell colony formation reported that immediately transfectd mi R-185 mimic could retrain the formation of MGC803 cell colony. The Flow Cytometry showed that:immediately transfectd mi R-185 mimic of MGC803 could promote the postpone of G1 period and cell apoptosis.The result of real-time PCR and western blot demonstrated that the level of immediately transfectd mi R-185 mimic decreased, compared with immediately transfectd Sh NC group, reversed Reagent in the control group and CDC42 in blank control group.Conclusions 1〠The expression of Mi R-185 is reduction in gastric cancer. 2〠Mi R-185 could restrain cell proliferation obviously and promote cell apoptosis. 3〠Mi R-185 could restrain gastric cancer cell line MGC803 proliferation and apoptosis by controlling CDC42 gene.
