Objectives:In SD rat liver model to research DNMT3 B and Akt1 expression and correlation to investigate whether Akt signaling pathway regulated DNMT3 B expression。Methods:1. The use of low-dose intermittent DENA carcinoma model established by intraperitoneal injection;Week 16 randomly selected cage model group rat to liver B ultrasound screening and stopped by intraperitoneal injection。The first 18 weeks all rats were sacrificed parallel pathology assessment modeling success,Edmondson method using tumor grade。2. The expression of DNMT3、Akt1 were detected by RT-PCR and immunohistochemistry method。3. Analysis DNMT3 B and Akt1 expression levels are correlated or not by linear regression statistical methods。Results:1. In the first 18 weeks, all rats were sacrificed(model group 12, control group 6), and visible liver nodules occupying;the pathological section shows cancer cells:two cases of well-differentiated,four cases of differentiated,six cases of poorly differentiated,the success rate ofcarcinogenesis in the model group is 70%。2. Detected by RT-PCR, the expression of DNMT3 B m RNA and Akt1 m RNA in model group was significantly higher than the control group(P<0.05); detected by immunohistochemistry, the expression of DNMT3 B and p-Akt in model group was higher than the control group(P<0.05)。3. The gene expression levels in the model group for linear regression study, found that the DNMT3 B 、p-Akt correlation coefficient was r=0.873,P=0.034,the DNMT3 B m RNA、Akt1 m RNA correlation coefficient was r=0.950, P=0.021 。Conclusions: DENA-induced liver cancer model in SD rats, DNMT3 B, Akt1 and phosphorylation levels showed high expression was positively correlated to participate in HCC。...
