Background:The glioma is the most common primary tumor, highly malignant, more invasive, high recurrence rate, short survival time, and low complete excision rate. It is a serious threat to human health. Recently, in the studies for the neural tumors, the glioma has never been overcome, and it is the difficulty and hot spot of clinical and basic researches.Glioma is the current focus in the study of tumor neurosurgery research field.The genetic molecular level of in-depth and practical research is particularly important.The glioma,involves anomaly of the structure and function of a variety of oncogene and tumor suppressor gene, is the abnormal expression of a multiple genes produces disease.Under the circumstances of the external stimuli activate and (or) enhance one or more of the original cancer gene expression, and internal with external factors for tumor suppressor gene mutating or (and) missing, cell within corresponding biochemical signal of turned guide pathway occurred disorders, it makes tumor cells far from normal adjustment mechanism, that made the tumor cells occur significantly proliferation and invasion.and showed the malignant biological behavior.In the current study, a large number of abnormal gene mutations and signaling pathways involved in tumor gene, that is still failed to study thoroughly.But the emergence of miRNA interpretation will be a key to explain the malignant tumor development process and the mechanism.MiRNA are exists in eukaryocyte and have evolved conservative tiny fragments of gens non-coding RNA, it involved in controlling cell differentiation, proliferation and programmed cell death, and other important pathophysiologic process.In today’s hot spot of the study showed that miRNA, in relation to the occurrence and development of tumor,has a significantly influence.MiRNA factor not only can be used as a tumor suppressor, restrain the activation of proto-oncogenes, also can be used as oncogene promote tumor growth, involved in the regulation of tumor occurrence, development, invasion and metastasis process, thus it can provide a new train of thought of diagnosis, prognosis and treatment of tumor.MiR-451 is a highly conserved micro RNA.Studies have reported that miR-451 has important relationship in the hepatopirtic system differentiation, neural development, cardiovascular disease, cancer and other disease processes, but also involved in the occurrence and development process of various kinds of gastric cancer, leukemia and other malignant tumors. Using miRNA as target to inhibit the growth and metastasis of tumor cells has become a new biological treatment strategies.And studies have begun to try to use chemical synthesis method to get the miR-451 oligonucleotide analogue for transfecting human brain glioma cells to inhibit the growth of tumor and invasive research purposes which won the new hope to the treatment of slioma.Research method:The current study only based on miR-451 with glioblastoma (grade IV gliomas) correlation research, whether the biological effects of the expression level and expression of universality and relativity in different grade gliomas are still needed further study. The purpose of the study is divided into the following several points:To explore the expression levels of miR-451 in different grade gliomas for discussing whether there is universality in different grade gliomas; MiR-451 in different grade gliomas represent differences,that is exploring whether it is of particular existence in different grade gliomas on miR-451 at different levels;To explore if there are some correlations for miR-451 in different grade gliomas represent with glioma cell differentiation degree and malignant glioma degree; To explore the glioma in clinical manifestation imaging, anatomy, pathology, molecular biology of the inner link and the possible mechanism. To further enrich the research of miR-451 and glioma, it has laid the foundation for the treatment of miR-451 in tumor early diagnosis and molecular levels.The research content:This topic research at all levels the expression level of miR-451 in glioma, and correlation of miR-451 of different level with different tumor invasive performance. Experiment is divided into two parts:collect in December 2013-December 2014 clinical glioma surgically resected specimens of 40 cases,part of which were practiced to HE staining, pathological classification, the other part were practiced to the PCR detection of tumor expression levels of miR-451. Through the statistical analysis methods to analyze the correlation between pathological classification and the expression of miR-451 levels.,for finding if there have linear correlation between them.Results:The expression of miR-451 and glioma grade has significant correlation. In low grade gliomas, the expression level of miR-451 is high. Along with the increase of malignancy, the expression level of miR-451 decreased gradually. This allows us to draw the conclusion that the content of miR-451 gradually reduce with the pathological grade of glioma rising.The research conclusion:1.According to the previous studies, miR-451 is most closely with invasion of spongioblastoma. This set of experiments, the high degree of malignancy of glioma have high invasion and low levels of miR-451 expression in glioma; low malignant degree one have the low invasion, and miR-451 expression level is high. Therefore, the expression of miR-451 is widespread in different grades of glioma tumors, and directly reflects the invasion and malignant degree of tumor. MiR-451 in the organization for human glioma cells express is generally lower.The current researches are treatments for some cell lines and spongioblastoma,so the universality of the expression characteristic will enlarge the scope of the present study to lay the theoretical basis and foundation.2.miR-451 expression in human glioma tissues was significantly different. At the same time, the difference of expressive content also as a feedback regulator regulates tumor cell, so that the glioma of different malignancy showed significant differences in the biological behavior of growth and metabolism, differentiation, migration, metastasis and vascular proliferation.3.The expression of miR-451 was the correspondence of the invasive glioma cell.Compositing of clinical imaging, anatomical and pathological basis of operation, invasion is the most prominent and most important characteristics of malignant glioma. Invasion,as a key, is closely associated with the relationship between them. It makes that miR-451 have important and far-reaching significance in the future in the field of research and treatment. The possible ways to increase miR-451 can fully inhibit the glioma cell invasive, in combination with other chemotherapy drugs, or other tumor gene regulatory molecules, or administered by transfection of cells or other means, which will bring ideas and new potential space for the treatment of glioma.4.MiR-451 in judgment of pathological results and the clinical prognosis has the potential value for further research. Whether different levels of glioma, or the same level of glioma, its content is lower, while the degree of malignant tumor is higher and the prognosis is worse. By finding the expression of different grade glioma miR-451 levels, formulating a reasonable scope and boundaries of reference values,and according to the content of miR-451,we can guide the glioma diagnosis decision and judgment of prognosis.
