Incidence of gastric cancer is a multi reason, many aspects of the process, including carcinogens such as Helicobacter pylori infection, tobacco, nitrates and nitrites and other reasons; Process involving multiple genetic and epigenetic changes, such as oncogenes, tumor suppressor genes, cell cycle regulation, cell adhesion molecules, DNA repair, genetic instability, abnormal telomerase activity. Recently, by chromatin instability induced by tumor heterogeneity concerns that chromatin instability and tumor resistance, relapse, is closely related to poor prognosis. Her-2 overexpression associated with chromatin instability in patients with gastric cancer in the biopsy, Her-2 overexpression, express higher levels of chromosomal instability, and gastric cancer and prognosis. Some studies have shown that gastric cancer when certain factors leading to AKT or Wnt signaling pathway β-catenin phosphorylation, can activate β-catenin and promotes tumor invasion and metastasis, now that chromatin instability related.Objective:This study was to detect gastric adenocarcinoma, metastatic lymph node tissue and adjacent tissues Her-2, β-catenin, and the expression levels of CIN, and to explore the depth of invasion, tumor stage, tumor differentiation, lymph node metastasis relevance, and the mutual influence and the role of Her-2, β-catenin, CIN between.Methods:First Affiliated Hospital of Yangtze University, surgical removal of the tumor and by two independent diagnostic pathology professor from September 2012 to January 2014 period for gastric adenocarcinoma tissue specimens were 156 cases of paraffin blocks, corresponding to 40 cases of metastatic lymph nodes and cancer 12 cases of adjacent tissues as the control group. HE staining evaluation of CIN in chromatin drawing number and immunohistochemical detection of SP Her-2, the expression levels of β-catenin, and analysis of the above indicators and gastric cancer infiltration depth, tumor stage, the correlation between the degree of differentiation, lymph node metastasis, and Her-2,β-catenin, the interaction between CIN.Results:In gastric adenocarcinoma strong expression of Her-2 was 15.4% (24/156), moderately positive expression rate of 18.0% (28/156), weakly positive expression rate of 19.2% (30/156), negative expression was 47.4% (74/156); in 40 cases of metastatic lymph nodes, the strong expression of Her-2 was 10.0% (4/40), moderately positive expression rate of 5.0% (2/40), weakly positive expression rate of 15.0% (6/40),70.0% (28/40)negative expression; in 12 cases in the control group, the strong expression was 8.3% (1/12), moderately positive expression rate of 0% (0/12), weakly positive expression rate of 25.0% (3/12), with negative expression was 66.7% (8/12). In gastric adenocarcinoma β-catenin expression was cell membrane of 29.5% (46/156), and the absence of abnormal expression rate 70.5% (110/156),40 cases of metastatic lymph nodes in the cell membrane expression was 10.0% (4/40), abnormal expression and lack of expression was 90.0% (36/40), β-catenin in 12 cases of the control group, cell membrane expression was 100%(12/12); CIN the positive expression rate was 44.9% (70/156), negative expression was 55.1% (86/156), the positive expression in 40 cases of metastatic lymph nodes was 35.0% (14/40), negative expression in metastatic lymph nodes was 65.0% (26/40), CIN in the control group expression was 16.7% (2/12),and 83.3% (10/12) negative expression. CIN, Her-2, p-catenin expression were associated with the patient’s age, gender irrelevant (p> 0.05), and tumor stage, infiltration, differentiation, lymph node metastasis; Gastric adenocarcinoma primary tumor tissue of Her-2 expression compared with lymph node metastasis was no significant difference p> 0.05;β-catenin membrane expression compared to metastatic lymph nodes was significant difference p<0.05,; there is no significant difference in the expression levels of CIN with lymph node-positive expression levels of CIN, p> 0.05. Gastric adenocarcinoma Her-2 and β-catenin expression was found by analyzing r= 0.407, p<0.05; Her-2 and CIN expression was found by analyzing r= 0.550, p<0.05; CIN and P-catenin expression was found by analyzing r= 0.442, p<0.05; in metastatic lymph node tissue, Her-2 and β-catenin expression was found by analyzing r=-0.195, p> 0.05; Her-2 and CIN expression was found by analyzing r= 0.039, p> 0.05; CIN and P-catenin expression was found by analyzing r= 0.301, p> 0.05 were not correlated.Conclusion:CIN, Her-2,β-catenin expression were associated with the patient’s age, sex not correlated with tumor stage, infiltration, differentiation, lymph node metastasis; Gastric adenocarcinoma primary tumor tissue of Her-2, no significant difference in the expression level of CIN expression with lymph node level; β-catenin membrane expression compared with lymph node difference statistical significance; in gastric adenocarcinoma Her-2, β-catenin, CIN expression correlation of can affect each other. Her-2, P-catenin, CIN can help determine the biological behavior of tumor malignancy, tumor invasion and metastasis ability. |
