Research Purposes:Recently, many research evidences show that inflammation and immune cells accumulate in the central nervous system, kidney and arteries, CD4+CD25+FOXP3+ regulatory T cell is a kind of subsets of lymphocytes, it has inhibiting effect, by adoptive transfer or removing by man can improve or deteriorate hypertension.So we speculate that immunolo--gic mechanism may has certain relationship with pathogenesis to essential hypertension. In order to know whether has differences between the essential hypertension group and the health group, how the immunologic mechanism effects essential hypertension, and offer new theoretical bases and strategies for the immunotherapy of hypertension, we study the number and function of CD4+CD25+Foxp3+Tregs in the peripheral blood.Research methods:According to the 2010 Chinese guidelines for the management of hypertension,peripheral blood samples were collected from 30 patients with level 2 to level 3 essential hypertension who were treated in department of cardiology of Changsha center hospital during July 2014 to Oct. 2014(15 men and 15 women), their average age is 67±12 years,and 30 healthy donor as control group(16 men and 14 women),their average age is 64±9 years.Collecting their basic clinical data, Lipid, blood glucose, peripheral blood lymphocyte count,hypertension history and hypertension therapeutic regimen.Taking elbow vein blood by heparin sodium anticoagulation tube, peripheral blood ratio of CD4+CD25+Foxp3+tregs were evaluated by flow cytometric analysis,FOXP3 m RNA in peripheral blood monocyte cells were determined by RT-PCR.Results:(1)The proportion of CD4+CD25+Foxp3+Tregs in the peripheral blood of the essential hypertension group is more less than the control group(essential hypertension group vs. control group, 4.49±1.30% vs. 5.29± 1.59%,P<0.05);(2)The expression of Foxp3 m RNA of PBMC in the peripheral blood of the essential hypertension group is obvious less than the control group(essential hypertension group vs.control group, 3.81±3.23 vs. 8.41±6.93,P<0.01).Conclusion:(1)Compared with the health control group, the number of CD4+CD25+Foxp3+Tregs is reduced and the express of it’s Foxp3 m RNA is decreased.These reminder that the decrease of the number and function of CD4+CD25+Foxp3+Tregs may have certain relationship with pathogenesis of essential hypertension.(2)By studying the biological characteristics and function of CD4+CD25+Foxp3+ tregs may find a new way to prevent and treat essential hypertension.(3)Proposing a hypothesis that immunologic mechanism may participate in the process of the occurrence and development of essential hypertension, the decreasing of the number and function of CD4+CD25+Foxp3+ tregs may take part in the activate of immunologic mecha-nism.
