Objective Using different concentrations of angiotensin Ⅱ(Ang Ⅱ) continuous stimulate the human umbilical vein endothelial cells(HUVECs) for 24 hours to simulate the condition of hypertension. To detect the expression of IL-33 and s ST2 in endothelial cells. To determine the changes of IL-33 and plasma s ST2 levels in patients with hypertension, and to explore the relationship between s ST2 and hypertension and confirmed that s ST2 affected the occurrence and to verify the correlation between the IL-33, s ST2 and hypertension.Methods A.The part of Cell experiment: The HUVECs were cultured and selected the 5-8th generation to be the experiment object.The research subgroups were as follows: The Ang Ⅱ 1 group, Ang Ⅱ 2 group, Ang 3 group and the control group. Cell concentration were control in 1 × 106/ml, the use of concentration of Ang Ⅱ with 0, 10-5mol/L, 10-3mol/L and 10-1mol/L to intervention and continue training 24 h. Using ELISA method to detect the IL-33 and s ST2 concentration in serum culture medium. B. All groups were recorded on the basic information,medical history, blood pressure measurement and venous blood. preserved in-80 C refrigerator after detect the serum lipid, the using ELISA method for the determination of IL-33 and s ST2 concentration in plasma.Results A.The part of Cell experiment:(1) In HUVECs study, Ang Ⅱ 1 group [(99.51 ± 51.19)pg/ml], Ang Ⅱ 2 group [(199.28 ±21.71)pg/ml] and Ang Ⅱ 3 group [(167.93 ± 6.98)pg/ml] medium concentration of IL-33, were higher than those in the control group [(42.42±0.83)pg/ml], there was statistically significant difference(P < 0.05); Ang Ⅱ 2 group medium concentration of IL-33, higher than that of Ang Ⅱ 1 group and Ang Ⅱ 3 group, statistically significant difference(P < 0.05); four groups of medium concentration of s ST2, Compared with each other were statistically significant(P < 0.05). B. The part of clinical trials:(1)In clinical study, hypertension group IL-33 [(311.18±411.96)pg/ml] and ST2 [50.14±50.85)pg/ml] concentration is higher than in the control group [(149.25±245.43)pg/ml] and [50.14±50.85)pg/ml], and the difference was statistically significant(P < 0.05).(2) Concentration of IL-33 grade 1 [(268.52±374.02)pg/ml], grade 2 [(259.31±342.22)pg/ml] and grade 3 [(405.72±500.33)pg/ml] were higher than those in the control group[(149.25±245.43)pg/ml], there was statistically significant difference(P < 0.05); grade 3 were higher than grade 1 or grade 2, there was statistical significance(P > 0.05).(3) Four grade of Plasma concentration of s ST2, Compared with each other were statistically significant(P < 0.05).(4) Concentration of IL-33 and value of systolic blood pressure(r=0.185, P < 0.05) and values of diastolic blood pressure(r=0.183, P < 0.05) has no obvious correlation; and concentration of IL-33 and value of mean arterial pressure(r=0.214, P < 0.05) were positively correlated;(5) Concentration of s ST2 and the value of systolic blood pressure(r = 0.211, P < 0.05) and the value of diastolic blood pressure(r = 0.327, P < 0.05) and value of mean arterial pressure(r=0.325, P < 0.05) were positively no correlated; between concentration of IL-33 and concentration of s ST2 has no obvious correlation(r = 0.133, P > 0.05).Conclusions 1. Inflammation involved in primary hypertention. IL-33 maybe involved in inflammation of primary hypertension. 2. The s ST2 may be involved in the process of occurrence and development of primary hypertension through the combination of competition with IL-33. 3. The s ST2 is expected to become a risk factor for hypertension.
