Objective To observe the expression and distribution of DCAMKL-1 and Ki-67 in pancreas after Partial pancreatectomy and to investigate the regulation mechanism of islet regeneration. Methods 35 adult SD rats were randomly divided into a normal control group(n=5), and a model group(n=30), which was further divided into 6 groups(n=5). The operation resection of pancreatic(about 60-70%) was performed after 1, 2, 3, 4, 5, and 6 d from the remnant pancreas, immunohistochemical and PV-9001 staining was used to observe the expression and distribution of DCAMKL-1 and Ki-67 in pancreatic tissues. Results DCAMKL-1 and Ki-67 had a little expression in the normal pancreatic tissues. The expression was mainly distributed in the duct, pancreatic islet and mesenchyme between acini. In the model group 1 d after operation, there were a large number of cells like honeycomb around the main pancreatic duct, and DCAMKL-1 and Ki-67 expression was significantly increased. After 3 d, cells increased obviously and the emergence of duct was like a structure around the major duct. DCAMKL-1 and Ki-67 had an expression peak. After 4 d, the expression began to decline, but was still higher than normal at 6 d. Compared with the normal group, the expression of DCAMKL-1 did not change significantly in the acini and islets, and Ki-67 had a peaked at 5 d, and declined at 6 d. Conclusion After partial pancreatectomy, cell proliferation is mainly in major pancreatic duct and present DCAMKL-1-positive, and DCAMKL-1+ cells may be as pancreatic stem cells.
