Objective:Study of Cervus elaphus Linnaeus in Talim Xinjiang main component of velvet antler polypeptides in different areas and different harvest period between content and activity difference, to provide a reference basis for reasonable development and utilization development of Xinjiang Tarim red deer standardized breeding and velvet antler medicinal materials. Methods:According to the 2010 version of Pharmacopoeia,identification of Cervus elaphus Linnaeus in Talim Xinjiang, and describe their properties.With glycine as control sample, the alcohol extract of velvet antler on TLC. The velvet antler of moisture, total ash and alcohol soluble extractive were determined.The velvet antler polypeptides extracted by the method of sodium acetate solution. The concentration of antler polypeptides from Cervus elaphus was measured by BCA Protein Assay Kit. The chemical composition was determination using sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE).MTT assay and trace enzyme standard method was used to detect the effect of antler polypeptides of mouse into the influence of activity of osteoblasts(MC3T3-E1) on the proliferation and alkaline phosphatase(ALP). Results:With the same harvest times of anlters were smaller; first born antlers are collecting beautiful velvet antler Sancha. Compared with the first crop of velvet, the smaller of the backbone of the second crop of velvet, ramose less.TLC of G-254 showed that 5 areas antlers and antler(red deer) control medicine appear the same main spots. The research shows that check in formulating moisture was less than 9%;tolal ashes were less than51%;alcohol soluble extraction was more than 2%. The results showed that BCA protein quantitative determination, there are differences in antler polypeptide content in different areas, there are also significant differences in the the content of pilose antler of different harvest time. Electrophoresis results showed that 5 areas antler polypeptide compositionsof electrophoresis were similar, it′s mainly bands showed 3 bands, respectively<40 KD, 25 KD, 15-10 KD. In vitro activity showed that velvet antler polypeptide has the effect on the proliferation of MC3T3-E1 cells and the secretion of ALP. From different areas and different harvest times of velvet antler polypeptides on MC3T3-E1 cells promoting proliferation and promoting alkaline phosphatase(ALP) secretion activity showed a difference(P<0.05). Concluison:The subject of the quality standard of antler was studied,the contents of 5 different areas of Xinjiang province antler polypeptide was determined,and the use of distribution. SDS-PAGE detection of velvet antler polypeptide protein components at the same time, the anti-osteoporosis bioactivity mechanism of antler polypeptide in vitro were studied. The experimental results for the Xinjiang Talim red deer standardized breeding and reasonable charge, and saw the development and reason utilization of antler to provide the experimental basis.
