Objective: To study the influence of blackcurrant seed powder on renal hypertensive rats blood pressure, heart, kidney, and to discuss blackcurrant seed powder antihypertensive mechanism. Methods: To prepare two kidney one clip hypertensive model, model rats which were built successfully were randomly divided into model group,captopril group(15mg/kg/d), bezoar step down group(0.208g/kg/d), blackcurrant seed powder low dose group(1.375g/kg/d), middle dose group(2.75 g/kg/d), and high dose group(5.5g/kg/d). Another 10 rats were divided as control group. Each group was treated by intragastric administration eight weeks. The control group, model group were given the same volume of distilled water. Blood pressure were measured in rats before and after treatment, and in the last treatment, abdominal aortic blood sampling measured thromboxaneB2(TXB2), 6-ketone-prostaglandinF1α(6-Keto-PGF1α), angiotensin II(AngII),endothelin(ET), nitric oxide(NO), malondialdehyde(MDA) levels and superoxide dismutase(SOD) activity; the heart and kidney were taken to measure cardiac index, left ventricle index, renal index. Results: After 8 weeks of treatment, to measure blood pressure, compared with model group, each dose group of blackcurrant seed powder could decrease renal hypertensive rats blood pressure(P<0.01); to measure cytokine in rats’ blood, compared with model group, each dose group of blackcurrant seed powder ET,TXB2, MDA levels were significantly lower(P<0.01 or P<0.05), NO level increased significantly(P<0.01), blackcurrant seed powder middle and high dose group AngII levels were obviously decreased(P<0.01 or P<0.05); blackcurrant seed powder high dose group6-Keto-PGF1α level and SOD activity were increased significantly(P<0.01 or P<0.05); to measure left ventricle index, cardiac index, compared with model group, each dose group of blackcurrant seed powder left ventricle index decreased significantly(P<0.01 orP<0.05), blackcurrant seed powder high dose group cardiac index decreased significantly(P<0.05). Conclusion: Blackcurrant seed powder can reduce the blood pressure of the renal hypertensive rats, and play a protective effect role on the heart, it is speculated that the step-down approach may be by protection endothelial cells through antioxidation, and regulating vasomotor factor to improve endothelial function at the same time.
