Objective:Discussed before and after neoadjuvant chemotherapy in cervical squamous carcinoma tissues miRNA-138 and miRNA-720 test their target protein expression differences and H2AX with EZH2 expression quantity before and after chemotherapy, analyzes its relation to chemotherapy curative effect, and explore the detection sensitivity of chemotherapy. Methods:Using quantitative (Q-RT) real-time PCR technology for 36 cases of cervical squamous carcinoma tissues before and after neoadjuvant chemotherapy in the expression of miRNA-138 and the miRNA-720, at the same time, application of Western blot technique to detect the target protein gamma H2AX (phosphorylation H2AX) with EZH2 expression. Results:Compared with the prior to neoadjuvant chemotherapy, chemotherapy group effective chemotherapy of miRNA-138 and the miRNA-720 expression were raised, the miRNA-138 and the miRNA-720 average expression differences before and after chemotherapy were 0.8801±0.1787 and 0.8801±0.0567, for chemotherapy before the chemotherapy after 0.89 times and 0.86 times, the difference had statistical significance (p<0.05);And the target protein gamma H2AX with EZH2 express lowered after chemotherapy before the chemotherapy, the average expression differences before and after chemotherapy were 0.6618 ±0.1626 and 0.6618±0.2367, for chemotherapy before the chemotherapy after 4.6 times and 5.4 times, the difference had statistical significance (p<0.05).Group and invalid miRNA-138 and the miRNA-720 average expression differences before and after chemotherapy was 1.5699±0.3776 and 0.3776±0.2625, the target protein gamma H2AX and EZH2 average expression differences before and after chemotherapy was 0.9856±0.1418,0.5415±0.1914, there were no statistically significant difference (p>0.05). Conclusion:The miRNA-138 and the miRNA-720 and its target protein is associated with cervical cancer chemotherapy curative effect, and the miRNA-138 and the miRNA-720 before and after chemotherapy by inhibiting the gamma H2AX with its target protein EZH2 expression of negative regulation, provide basis for selection of chemotherapy for cervical cancer patients undergoing chemotherapy sensitivity index, is easy to individualized treatment of tumor.
