Studies On Chemotherapy Effects Of Gemcitabine And Caboplatin On Human Ovarian Cancer Cell Line SKOV3 And Tumor-Bearing Nude Mice

Objective:1) Effects of different groups GEM and Carbo on apoptosis induction of SKOV3 cells2) Inhibitions of different groups of GEM and Carbo on cell- cycle phase of SKOV3 cell lines; Immunofluorescence staining of CDK2 proteins on SKOV3 cells after treated with different drugs.3) Tumors from SKOV3 cells were established by subcutaneously in nude mice. The inhibition effects of different drug groups on tumor growth were studied; the inhibition effects on tumors newly formed blood capillaries treated by different drug groups were shown by immunohistochemisty.4) Methods: MTT detected growth-inhibitory effect of different concentrations of GEM and Carbo on SKOV3 cell lines. MTT detected growth-inhibitory effect of low concentrations of Carbo combined with GEM on SKOV3 cell lines; FCM detected GEM and Carbo used only or together in cell-cycle phase or apoptosis induction of SKOV3 cell lines. Immunofluorescence staining of CDK2 proteins on SKOV3 cells after treated with different drugs.Construct SKOV3 tumorbearing nude mice. After the tumor model was constructed successfully, they were divided into GEM group, Carbo group and Carbo combined with GEM group. They were injected subcutaneously alternate-day and to last 3 weeks. The tumor volumes in different days were watched.Results1) GEM and Carbo can inhibit cell proliferation in certain concentrations and have dose-dependent character. GEM in 4 μM can inhibit cell proliferation dramatically and for Carbo, this concentration should be increased to 100μM can reach same inhibitory effects. However, if Carbo combined with GEM, then 20μM plus 5μM Carbo can inhibit cell proliferation dramatically.2) GEM decreased cell numbers of S phase mainly, and Carbo haven’t this result after treated with the same concentrations but the apoptosis peak appeared before the G1 phase; GEM combined with Carbo inhibited cell division totally, the numbers of cell in S phase was zero.3) Different groups of GEM and Carbo all have apparently apoptosis induction effect; the percentages of different groups haven’t apparently difference.4) Immunofluorescence staining of CDK2 proteins showed that different treatment inhibited CDK2 expression to some extent.5) In vivo experiment of tumor-bearing nude mice showed that Carbo itself or Carbo combined with GEM have a good inhibitory effect on tumor growth; Immunohischemistry of CD31 showed that the newly formed blood vessels of Carbo combined with GEM decreased.Conclusion: GEM can be used with Carbo. In this situation, this two drugs can exert a positive effect in very low dose and decreased the newly formed blood vessels in xenograft nude mice.
