| Objective:Investigation of the expression of cancer stem cells(CSC) markers of ABCG2 〠CD44 〠CD34 in human nasopharyngeal carcinoma(NPC) CNE-2 〠5-8F 〠and 6-10 B cell line,and the Clone-forming capability of ABCG2+ã€CD44- and CD34+ cells of 5-8F. to choice a possible cancer stem cell marker for the research of NPC.Methods:Flow cytometry was used to investigate the expression of cancer stem cells of markers ABCG2ã€CD44ã€CD34 in CNE-2ã€5-8Fã€and 6-10 B. Clone-forming capability of ABCG2+ã€CD44- and CD34+ cells of 5-8F was determined by using softer agar clone formation assay.Results:ABCG2 positive cells accounted 0.1%ã€18.6%ã€19.9% in 5-8F ã€6-10 B ã€CNE-2 cell lines;CD44 positive cells accounted 99.5%ã€93.2%ã€99.1% in 5-8F 〠6-10 B ã€CNE-2 cell lines;CD34 positive cells accounted 0.0 % 〠3.0 % 〠0.1 % in 5-8F 〠6-10 B 〠CNE-2 cell lines;Compared with ABCG2- cells, ABCG2+ cells show higher clone-forming capability. CD34+ cells also have higher clone-forming capability then CD34- cells. There is no difference about the clone-forming capability between CD44+ cells and CD44- cells.Conclusions:1ã€The expression of cancer stem cells markers of ABCG2ã€CD44ã€CD34 is vary in different NPC cell lines. 2ã€ABCG2+ã€CD34+ cells of 5-8F have higher clone-forming capability then the ABCG2-ã€CD34-cells. 3ã€There is no difference about the clone-forming capability between CD44+ cells and CD44- cells. |
