Objective: Study on correlation between antibiotics resistance and virulence genes in Klebsiella pneumoniae isolates, through analyzing the drug resistance spectrum and distribution of virulence gene of clinical isolates of klebsiella pneumoniae, which provides the theoretical foundation for clinical treatment.At the same time study on homology analysis of klebsiella pneumoniae, providing the phylogenetic relationship in variation, differentiation, propagation law among strains.Methods: Collect 119 strains of pneumonia klebsiella isolates, and use PCR technique to detect resistance gene SHV, TEM, CTX-M, OXA, and screen strains of ESBLs strains. Drug sensitive slips method is used to analysis its drug resistance. Use PCR technique to detect seven kinds of virulence gene rmp A, aer- 1, aer- 2, mag A, gyrb, ybt, wca G, and compare virulence genes distribution between ESBLs and not ESBLs pneumonia klebsiella strains, and analyse the correlation between multiple drug resistance and virulence genes distribution.Though Bio Edit software editing pieces of the same number of sites, use MEGA- 4 to cluster phylogenetic tree as homology analysis of strains.Results: Screened drug-resistant genes SHV, TEM, CTX- M, OXA in 119 strains of pneumonia klebsiella isolates, and selected 32 strains of ESBLs pneumonia klebsiella. Though drug-resistant spectrum analysis to 119 strains klebsiella pneumonia,we can see above three and multiple resistance to antimicrobial agents can reach 41.17%, 8 to 12 kinds of antibiotics resistant strains accounted for 30.25%. Different virulence gene detection rate differences were statistically significant(P < 0.05). 2 to 5 kinds of different virulence genes strains are relatively high, above 15%.Comparing the not ESBLs of pneumonia klebsiella,the rate of distribution of virulence genes of ESBLs of pneumonia klebsiella is much lower. For multiple resistance isolates and virulence gene number correlation analysis, the correlation coefficient between virulence gene distribution and drug resistance is 0.227(P < 0.05), negatively correlated, which suggests that the more number of distribution of virulence genes of strains, the chances of getting multiple drug resistance phenomenon is smaller. The correlation coefficient between distribution of virulence genes and resistance genes is 0.195(P < 0.05), negatively correlated, that is to say, the more number of distribution of virulence genes of strains, the less chances of getting drug resistance gene.With MEGA-4 to make phylogenetic tree of A, B, C, D, E, F six group, through the chart,we can see A group is the advantage group, followed by B group. ESBLs in group A were given priority to the ESBLs of other groups. In group A, virulence genes of ESBLs contains less relative to other group of virulence genes of ESBLs, through the corresponding to the front of the relationship between the resistance and virulence, can be found that B, C, D, E, F group are more virulence genes, A group of virulence genes is less. Homology between the flora is higher, there is a clone of the outbreak. Conclusion: There are correlation between virulence genes distribution and the drug resistance of klebsiella pneumonia, inclined to negative correlation.Virulence gene distribution phenomenon in the more number of isolates, the probability of multiple drug resistance is small, which providing reasonable clinical treatment and some theory basis.
