| Objective:To investigate the neuroprotective effects of Ginsenoside Rg1 in a rat traumatic spinal cord injury(SCI) model,and analyze the probable mechanism of its protective treatment role.Methods: forty-five rats,weighted 220~260g,were randomly divided as three groups:group A(n=15),group C(n=15),each rat received injection with the same amount of Hydrogen Saline, and group B(n=15), each rats received injection of normal saline(NS).The rats of all group were assigned to A1(3d)ã€A2(7d)ã€A3(14d);B1(3d)ã€B2(7d)ã€B3(14d) and C1(3d)ã€C2(7d)ã€C3(14d),at the time 3days,7days,14 days after SCI. All the rats of Bã€C group were established model of spinal cord injury by the improved Allen’S collision method,and all the rats of A group were treated with the vertebral plate opened only.At the corresponding time,the BBB score of all rats were enforced,the content of Spinal cord tissue were detected with HE staining for pathological observation,detected the expression of GFAP with immunohistochemistry, detected the expression of Caspase-3 and Bcl-2 by western blot。 Before the rat were killed at each time point,the blood were exsanguinated for detection of MDA and SOD.The data were analyzed using statistical software SPSS 16.0.Results:1. BBB scale : The score of all subgroup in A group is 21 points;After 3 days, 7 days and 14 days,the BBB score of B group was higher than that of C group at appropriate times(P<0.05).2.HE staining:The cell morphology were intact,arranged compactly,and the nuclei were hyperchromatic in the A group.Compared with group B1,the inflammatory cell infiltration of group C1 was more evident; compared with group C3,the Neuron atrophy of group B4 was significantly milder;and the connective tissue formation of group C3 was significantly more than group B3.4. GFAP: the ratio of GFAP area of C group was higher than B group.5.The biological activity assay of MDA and SOD:MDA After 3 days, 7 days and 14 days,the level of MDA of C group was higher than that of B group at appropriate times.SOD The level of SOD was gradually reduced after SCI. After 3 days, 7 days and 14 days,the level of SOD of B group was higher than that of C group at appropriate times.6. The expression of caspase-3 and bcl-2Caspase-3 At the time of 14 days, the express of caspase-3 of C group was higher than that of B group(P<0.05). And the expression of caspase-3 of B group was higher than that of A group.Bcl-2 At the time of 14 days, the express of expression of bcl-2 of B group was higher than that of C group(P<0.05).Conclusion:1.Ginsenoside Rg1 have protective effect on spinal cord injury and it can improve the limb motor function recovery.2. Ginsenoside Rg1 can reduce the oxidative stress after spinal cord injury,inhibit the neural apoptosis, slow expression of GFAP, thus reducing formation of glial scar, promote recovery of nerve function after spinal cord injury. |
