Objective The present study investigated the effects of selective cyclooxygenase-2 inhibitor Celecoxib proliferation and apoptosis of cultured human renal carcinoma cell line OSRC-2 in vitro. Tumor growth inhibition of selective cyclooxygenase-2 inhibitor celecoxib in NOD-Scid mice.Methods The proliferation of OSRC-2 cells was studied by MTT assay.Western blot was used to detect the expressions of COX-2 of OSRC-2 cells.The expression of COX-2 m RNA of OSRC-2 cells was analyzed by reverse transcription-polymerase chain raction(RT-PCR).Flow cytometric analysis was used to detect the apoptosis status of OSRC-2 cells.A NOD-Scid mice model of renal carcinoma was reproduced.The effect of COX-2 inhibitor celebret on tumor growth was observed.Result Celecoxib can inhibit proliferation of renal carcinoma cell line OSRC-2 and induce its apoptosis in vitro.RT-PCR and Western blot showed the expressions of COX-2 were significantly inhibited by celecoxib.The expression level of COX-2 reduced and the weight and volume of the xenografts in NOD-Scid mice decreased more in the groups treated with celecoxib.Conclusion The result suggesting that the celecoxib could effectively suppress the growth of OSRC-2 cells,the mechanism probably relates to its inhabited expression of COX-2 and induce apoptosis.COX-2 may have participated in the course of growth of xenografts and selective COX-2 inhibitor Celecoxib can inhibit the proliferation of transplantation tumor of renal carcinoma effectively.
