Objective: The experiment of anti-ulcer effect in vivo: Firstly, we maked three different kinds of gastric ulcer animal models in SD rats: pyloric ligation, indomethacin stimulation, water stress to contrast the therapeutic action of these three drugs on three different kinds of gastric ulcer animal models.The experiment of enzyme activity inhibition in vivo and in vitro: To compare the effect of dexrabeprazole sodium、rabeprazole sodium and S(-)-isomer sodium for injection on H+、K+-ATPase activity by giving drugs in vivo(water stress gastric ulcer in SD rats) and in vitro(normal SD rats).By the experiment of anti-ulcer effect in vivo and the enzyme activity inhibition in vivo and in vitro, we were to supplement pharmacodynamics research dates of dexrabeprazole sodium for injection and provide a reference basis for its listing in domestic research. Methods: The experiment of anti-ulcer effect in vivo: We established three different kinds of gastric ulcer animal models in SD rats: pyloric ligation, indomethacin stimulation, water stress, and then gave dexrabeprazole sodium for injection(dexpure-iv)、rabeprazole sodium for injection and S(-)-isomer sodium for injection through the tail vein。Respectively, we designed that the dose were 1 mg·kg-1、2 mg·kg-1、4 mg·kg-1 three dose groups, counted the ulcer index or ulcer scores of all groups, calculated the percentage by ulcer inhibition. By doing all those, we could observe contrastively how dexrabeprazole sodium for injection and rabeprazole sodium and S(-)-isomer sodium for injection were different about the prevention、 treatment effect on anti-ulcer activity of dexpure-iv in SD rats of those three different kinds of gastric ulcer animal models.The experiment of enzyme activity inhibition in vivo and in vitro: In vivo, we collected gastric mucosa parietal cell in SD rats of water stress gastric ulcer model by dissecting rats, and made of 2% gastric mucosa homogenate with normal saline after blending. According to the ELISA biometric kit instructions of the H+、K+-ATPase,we conducted the H+、K+-ATPase activity determination of gastric mucosal parietal cell to contrast the effect on H+、K+-ATPase activity among dexrabeprazole sodium for injection、rabeprazole sodium for injection and S(-)-isomer sodium for injection(iv). In vitro, we collected gastric mucosa of normal SD rats and mixed with normal saline to make 2% gastric mucosa homogenate. Then we blended gastric mucosa homogenate of eight SD rats as ELISA biometric kit instructions of the H+、K+-ATPase was said. There were some different operations that we joined diluent of dexrabeprazole sodium for injection with rabeprazole sodium and S(-)-isomer sodium for injection,the concentration gradient(unit:g·m L-1) was: 0、 1.7×10-9、 5.2×10-9、1.7×10-8、5.2×10-8、1.7×10-7、5.2×10-7,observing contrastively the inhibition of enzyme activity for dexrabeprazole sodium for injection、rabeprazole sodium for injection and S(-)-isomer sodium for injection in vitro.Results and conclusion: The experiment of anti-ulcer effect in vivo: We could become conscious of that the gastric ulcer model SD rats of pyloric ligation stomach had different damage visibly and visible to the naked eye, dispersedly or intensively they were in distribution with dark brown dot or sheet bleeding spots; the ulcer penetrated to muscular layer; the stomach mucosa ruffle reduced or even disappeared, appearing on translucent or redness, having punch phenomenon seriously. There also had different damage in glandular stomach of gastric ulcer model SD rats of indomethacin stimulation and water stress with a dot or dark brown or yellow strip bleeding spots. Compared with model group, there had obvious inhibitory effect of dexrabeprazole sodium for injection to the three kinds of gastric ulcer animal models in SD rats(P < 0.01 or P < 0.05), better than rabeprazole sodium for injection and S(-)-isomer sodium for injection of the same dosage, which could significantly inhibit the occurrence of gastric ulcer.The experiment of enzyme activity inhibition in vivo and in vitro: There was nosignificant difference among dexrabeprazole sodium for injection、rabeprazole sodium for injection and S(-)-isomer sodium for injection in inhibition of H+、K+-ATPase activity in vitro. However, compared with model group, all these three drugs had inhibitory effect on H+、K+-ATPase activity in water stress of those three drug doses of 1 mg·kg-1、2 mg·kg-1、4 mg·kg-1. It could be back to H+、K+-ATPase activity level of normal SD rats in high doses 4 mg·kg-1, and the inhibitory effect was stronger than rabeprazole sodium for injection and S(-)-isomer sodium for injection.
