Experimental Studies Of The Affects Of Banqiao Dangshen Polysaccharide On Expression Of SOD, MDA, IL-β, MMP-13 In Osteoarthritis Chondrocyte

Objective:To observe the affects of Banqiao Dangshen Polysaccharide(BDP) on proliferation of and expression of SOD, MDA, IL-1β,MMP-13 in chondrocytes from patients with osteoarthritis.Methods: Extract BDP with water extraction and ethanol precipitation according to the methods of Ultrasound-water extraction and alcohol precipitation. Separate chondrocytes from knee joint cartilage from patients with OA after total knee arthroplasty, digested by type II collagenase, and cultured with medium in Incubator above 37 5%CO℃2. Find out the suitable concentration and stimulating time of BDP in cultured P2 chondrocytes by MTT method. Divide cultured chondrocytes into 5 groups and co-cututred 48 hours with different concentration of BDP(low, middle, high does groups) or glucosamine or blank control respectively. Measure the activity of SOD with WST-1 method, and the concentration of MDA and IL-1β in the cells with ELISA method, the expression of MMP-13 with Immunocytochemical method.Results: The refining degree of BDP was 91.09±1.19% with producing rate of 5.25%. BDP significantly increased the proliferation of osteoarthritic chondrocytes with dose-dependent effect. No affects of BDP on expression of SOD were observed in different dose of stimuli(P>0.05). BDP with middle and high dose reduced the level of MDA(P<0.05, P<0.01, respectively). However, there was no significant difference between BDP groups control(P>0.05, respectively). BDP reduced the level of IL-1βwith dose-dependent effect(P < 0.01, respectively). BDP down-regulated the expression of MMP-13, which shows significant differences compared with blank control group.Conclusion:BDP could increase the proliferation of osteoarthritic chondrocytes and down-regulate expression of MDA, IL-1β and MMP-13. The protective role of BDP should be further investigated.
