| Obesity is an increasingly serious problem at a global scale, which is called global pandemic and has become an important public health and medical problems in twenty-first century. There are more than 1 billion adults and 10% of children are overweight or obesity all over the world. In China, obese population had amounted to 325 million in 2010 according to the people’s daily online, which was more than America, Britain and Australia, moreover, the figure could double in the next 20 years. More notably, China obese children increased 28 times in 15 years and nearly one-fifth overweight children were below 7 years old according to statistics, the proportion was far more than the European countries. Besides, Obesity can lead to other related diseases and has become the sixth-largest disease risk factors in the world, it can increase the morbidity and mortality of hypertension, diabetes, dyslipidemia, coronary heart disease, cancer and other disease. In Britain, the number of deaths from obesity is about 30000 people a year and 112000 to 280000 in USA. In addition, medical expenses on obesity have to be increased, and obesity will caused severe health problems, mental problems and economic burden. Hence, it is important to develop safe and effective weight loss drugs.Natural herbs are various, which are concerned by the pharmaceutical scientists increasingly on account of less toxic side effect and low cost in the treatment of diseases.Pancreatic lipase, plays a key role in the efficient digestion of triglycerides, is responsible for the hydrolysis of 50-70% of total dietary fats and can catalyze the hydrolysis of fats to fatty acids and glycerol which will be absorbed, However, if a drug can block decomposition of pancreatic lipase against fat, the absorption of fats for the body will be reduced and obesity can be prevented.In this paper, we studied Forsythia suspensa leaves’ effects on weight loss. The contents are as follows:First, the processed F. suspensa leaves (boiled 4-6 min) were extracted with distilled water, and followed by extracted systematically to obtain each sample. Second, spectrum efficiency relationship was analyzed between samples in order to infer the active ingredients in samples. Again, the effects of extracts on PL were measured by PNPB assays and the types of inhibition were determined by double-reciprocal plot. At last, animal experiment was done. The conclusions as follows:1. The extracts of F. suspensa leaves could inhibit PL. After extracting systematically, each extract exhibited different inhibitory effects on PL, the order from strong to weak were n-butanol phase, chloroform phase, ethyl acetate phase and crude extract. Their IC50 values were 1395.95,1487.37, 1769.29 and 2148.14 μg/mL respectively. The inhibitory effects had significant concentration-dependent relationships. Besides, crude extract and n-butanol phase suggested a competitive mode of inhibition against PL with respect to substrate concentration.2. N-butanol phase and crude extract were selected for animal experiment, the two substances reduced weight of obese rats, decreased total lipid content and triglyceride in serum. The results indicated n-butanol phase and crude extract had significant effect on weight loss and auxiliary function of reducing blood lipid.3. HPLC analysis of extracts of F. suspensa leaves indicated that the content of Forsythiaside A and Rutin in extracts was obviously positive correlation with their effects on PL, and the two exhibited different inhibitory effects on PL. The IC50 were 931.82 and 346.11 μg/mL respectively. The results showed that the inhibition effects of extracts on PL were related with Forsythiaside A and Rutin. Besides, the two components suggested a competitive mode of inhibition against PL with respect to substrate concentration. At last, spectrum efficiency relationship analysis to find the active substance is an effective and feasible method. |
