Objective:To study the effects of acupuncture,enriched rehabilitation training and acupuncture combined with enriched rehabilitation training on the motor function and the regulation of nerve regeneration inhibiting factor(Nogo-A) and Nogo receptors(Ng R) in rats after focal cerebral ischemia injury,so as to explore the effect and mechanism of acupuncture combined with enriched rehabilitation training on ischemic brain damage nerve functional recovery of curative effect than single treatment differences and their respective advantages.From the view point of regulation nerve regeneration factor regulating the action mechanism of the nerve regeneration, provide experimental foundation and theoretical basis for traditional rehabilitation therapy and modern rehabilitation clinical selective. Method:Rat models of local cerebral ischemia-reperfusion were established by threadoccluding the middle cerebral artery(MCAO).4 h after the surgery,Select 5 neurologic deficits 4-point scale of 1 to 3 points of ratsto divided into 5 groups:ham-operated group;model group;acupuncture treatment group;enriched rehabilitation training group; and acupuncture combined with enriched rehabilitation training group. The marks of neural function defect, sensation and motor ability were noted down separately. At 3d, 7d and 14 d of reperfusion,immunohistochemistry staining was applied to identify Nogo-A and Ng R cells in rats. The expression of positive cells in rats’ brain tissue were analysed among goups.Each group was divided into 3 subgroups by 3d,7d and 14 d.The marks of neural function defect, and the motor ability were noted down separately.At 3d,7dand14 d of cerebral ischemia injury,using HE staining observation ischemic infarction rats hippocampal tissue pathology change,the immunohistochemistry staining and western blot were applied to observe the expression of Nogo-A and Ng R cells in rats. The expression of Nogo-A and Ng R were analysed after cerebral ischemia among the above groups. Results:1 neural function defect scale:Sham operated group was normal,neuronal function evaluation of rats in another 4 groups had no obvious difference before therapy(P>0.05).3d,7d,14 d model and three treatment groups neural function defect scale gradually reduced after treatment;In 7d after treatment,the three treatment groups had lower scores,scores in acupuncture treatment group and enriched rehabilitation training group were remarkable(P<0.05)compared with model group,acupuncture combined with enriched rehabilitation training group was significant(P<0.01),acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.05).In 14 d after treatment,the three treatment groups had lower scores,scores in acupuncture treatment group and enriched rehabilitation training group were remarkable( P<0.05),compared with model group,acupuncture combined with enriched rehabilitation training group was significant(P<0.01),scores comparison between three treatment group, acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.01).2 sensory and motor function score:Evaluation results of rotating bar:3d,7d,14 d model and three treatment groups neural function defect training group were remarkable(P<0.05),compared with model group,acupuncture combined with enriched rehabilitation training group was significant(P<0.01),scores comparison between three treatment group,no significant differences between acupuncture treatment group and enriched rehabilitation training group(P<0.01).acupuncture combined with enriched rehabilitation training group has lower score was significant(P<0.01),acupuncture combined with enriched rehabilitation training group has lower score compared with enriched rehabilitation training group(P<0.01),acupuncture treatment group has lower score compared with enriched rehabilitation training group(P<0.01);Evaluation results of net screen: In 7d after treatment,the three treatment groups had lower scores,scores in enriched rehabilitation training group was remarkable(P<0.05)compared with model group,acupuncture combined with enriched rehabilitation training group was significant(P<0.01).Three score compared combined with enriched rehabilitation training group was significant(P<0.01),scores comparison between three treatment group,acupuncture combined with enriched rehabilitation training group has lower scores,the difference in comparison with the acupuncture treatment group was significant(P<0.01),acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture treatment group(P<0.01), enriched rehabilitation training group has lower score compared with acupuncture treatment grou P(P<0.01);3 The expression of Nogo-A Protein and Ng R:After cerebral ischemia injury,and sham operated group has not the obvious Nogo-A positive expression and the rest of the four groups in cerebral ischemia side hippocampus Nogo-A protein expression and Ng R has strong degree,around the brain ischemia model group Nogo-A expression of hippocampus were significantly higher than that of sham operated group,has the extremely significant difference(P<0.01).In 3d after treatment,Nogo-A protein expression and Ng R 4 group were increased regularity,three treatment group compared with model group,expression has no obvious difference(P>0.05);The expression of Nogo-A Protein:In 7d after treatment,acupuncture treatment group and enriched rehabilitation training group were remarkable(P<0.05)compared with model group,acupuncture combined with enriched rehabilitation training group was significant(P<0.01),scores comparison between three treatment group,acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.05),no significant differences between acupuncture treatment group and enriched rehabilitation training group(P>0.05);In 14 d after treatment,Nogo-A positive expression,acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.01),no significant differences between acupuncture treatment group and enriched rehabilitation training group(P>0.05);The expression of Ng R:In 7d after treatment,Three treatment group protein expression was extremely significant difference(P<0.01)compared with model group, scores comparison between three treatment group,acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.05),no significant differences between acupuncture treatment group and enriched rehabilitation training group(P>0.05);In 14 d after treatment,Ng R positive expression,acupuncture combined with enriched rehabilitation training group has lower score compared with acupuncture combined with enriched rehabilitation training group(P<0.01),no significant differences between acupuncture treatment group and enriched rehabilitation training group(P>0.05); Conclusion:1 The therapy of acupuncture treatment group and enriched rehabilitation training can enhance the recovery of sensory and motor functions by improving rats’ cerebral ischemia infarction area,signs and pathological damage and nerve function defect, Repair the nerve,and acupuncture treatment group and enriched rehabilitation training is superior to the simple acupuncture treatment and rehabilitation training;2 The therapy of acupuncture treatment and enriched rehabilitation training have their respective advantages in improving cerebral ischemia rats’ sensory and motor function,acupuncture treatment is good at improving the early feeling function, enriched rehabilitation training increased thigh muscle strength, improved walking, ability of cerebral infarction in rats,this may be related to two simple therapy is associated with nerve regeneration to inhibit the action of the factor;3 The therapy of acupuncture treatment group and enriched rehabilitation training can promote the effect of nerve functional recovery, may be related to inhibition of nerve regeneration inhibitory factor Nogo-A protein and Ng R expression.
