Objective This study was aimed to investigate the effect of emodin combined with 3’-azido-2’,3-dideoxythymidine (AZT) on the proliferation and apoptosis of the concentration of leukemia stem cells (CLSC) of KG-la cells.Methods The leukemia stem cells were acquired from human leukemia KG-1a cell lines by enriching tumor stem cell-like subpopulation with 5-FU, The CD34+CD38-subpopulation in KG-la cells were detected with flow cytometry, The effect of cell proliferation was detected by MTT assay, Cell apoptosis was detected by Flow Cytometry, The expression of NF-κB, BCL-2 and TGF-P mRNA and proteins were measured with RT-PCR and Western Blot.Results1. The emodin, AZT or the combined of them which could inhibit significantly proliferation of CLSC and appeared a time and dose dependent manner compared to the control group (p<0.01). The emodin’s IC50 was (244.1±0.84)μmol/L for 72 hours.2. The inhibitory rates on proliferation of CLSC treated by emodin combined with AZT for 72h is significantly higher than emodin or AZT single group, respectively, The synergistic inhibition effect is expressed after combined two drugs (CI<1).3. The apoptosis rates were (30.35±0.4)%, (39.35±0.43)% treated by fixed final emodin alone of 20 μmol/L combined with different concentrations of AZT of 2000μmol/L and 4000μmol/L for 72h,compared with the control group, the difference was statistically significant (p<0.05).4. Compared with cells treated with emodin and AZT drugs alone, the expression of NF-κB, BCL-2 and TGF-P mRNA and proteins decreased (p<0.05).Conclusion Emodin combined with AZT can synergistically inhibit the proliferation and induce cell apoptosis of CLSC. The possible mechanism of synergistic effect may be related with down-regulation of the expression of NF-κB, BCL-2 and TGF-β.
