Objective:Clear Tao Hong Si Wu Decoction (THSWD) on ischemic stroke tolerance and the neurovascular unit injury prevention and treatment. Will eventually provide a new theoretical basis for clinical prevention and treatment of stroke for THSWD is used, with important theoretical significance and application value.Methods:To temporarily blocking middle cerebral artery for establishment of a rat model of ischemic stroke (MCAO). Firstly, through the determination of the volume of cerebral infarction, neural function defect score, effects of ischemia and 2 hours later, different doses of Taohong Siwu Decoction pretreatment on ischemic tolerance; then through the determination of brain edema and the permeability of blood brain barrier, combined with the analysis, Western blot and immunohistochemical staining, immunofluorescence method to detect the vascular basement membrane protein Collagen Ⅳ, tight junction proteins occludin and Zonula occludens 1 (ZO-1) expression, so as to clear the Taohong Siwu Decoction pretreatment on ischemic stroke of neurovascular unit damage and protection. Finally, by Western blot and immunohistochemical detection of heme oxygenase-1 (HO-1) induced expression, explore the potential molecular mechanism of Taohong Siwu Decoction on prevention and treatment of the neurovascular unit damage.Results:THSWD can improve the tolerance of ischemic stroke in rats:compared with sham operation group, model group animal MCAO neural function damage score was significantly decreased, indicated that the model was successfully constructed, and compared with the model group, THSWD extract 0.5、1.0、1.5g·kg-1·day-1 group of animals the nerve function score increased significantly, suggesting that THSWD extract on ischemia reperfusion induced brain injury in rats has a protective effect. The volume of cerebral infarction was measured by TTC staining showed that compared with sham operation group, MCAO model group rats appeared obvious injury of cerebral infarction, show that the model successfully. And THSWD extract 0.5、1.0、1.5g·kg-1·day-1 group of cerebral infarction volume was significantly lower than that of model group rats. At the same time that the best dose of 1.0g·kg-1·day-1 effect of THSWD. The injury has a preventive effect of THSWD on neurovascular unit:compared with the model group, THSWD pretreatment group can significantly reduce the cerebral ischemia and cerebral edema after reperfusion, reduce the cycle of Evans blue into the brain, further studies show that the THSWD can increase the vascular basement membrane protein Collagen and tight junction protein expression the expression of occludin and ZO-1, protect blood brain barrier function, reduce the brain damage caused by ischemia reperfusion. Increased expression of Taohong Siwu Decoction can induce HO-1, compared with the model group, THSWD extract 0.5、1.0、1.5g·kg-1·day-1 group rats cerebral cortex HO-1 expression amount and the positive cells were significantly increased.Conclusion:In this experiment, found in the ischemia reperfusion model,THSWD showed significant neuroprotective effect. In our experimental results show that, before operation,THSWD by intragastric administration pretreatment can significantly improve the neurological function compared with the model, reduce the volume of cerebral infarction, cerebral edema, and can protect blood brain barrier permeability significantly. The effect may be through Nrf2 signal pathway.
