| Human β defensin-2 (hBD2) is a endogenous antimicrobial peptides which is expression widely on the human epithelial tissue has a bacterial function effectively,and the immune response,chemotaxis,protein synergistic effects are shown as well.However,the preceding research in human defensins function is always focus on the antimicrobial,the influence on cancer is still fuzzy.In the mean time.the gastric cancer is second leading cauce of cancers in the world.Even have some traditional treatments,the cancer cells migration,invasion and recurrence are obstacles for treatments as usual.Apparently,to find a safety and efficiency new therapeutic method for the gastric cancer is critically needed.Objective:Recombined human β-defensin 2 gene eukaryotic expression plasmid pCMV-hBD2,transient transfection of recombinant plasmid were to gastric cancer cell line BGC823 and normal gastric mucosal cell line GES-1.Finally,to detected the anti-tumor fuction on the gastric cells.Method:To recombined the human β-defensin 2 gene eukaryotic expression plasmid pCMV-hBD2 by using molecular biology way.Doing transient transfected to gastric cancer cell line BGC823 and normal gastric mucosal cell line GES-1 with recombinant plasmid.At the transient expression period,tested the expression on the mRNA and protein levels,according to done the MTS,colony formation,Transwell chamber experiments to detected the influnces on grastric cell line proliferation,colony formation,migration and invasion.Meanwhile,to detected the other related protein expression.Results:Using Gene sequencing to confirmed the hBD2 gene eukaryotic expression plasmid pCMV-hBD2 successfully.The hBD2 gene transfected cells line the relative expression of hBD2 was higher than others.The transfected hBD2 gene BGC823 cells line,the activities of proliferation,migration and invasion have became lower,but the GES-1 cells line proliferation is same as others.The transfected hBD2 gene cells line the relative expression of MMP2 was lower than others.Conclusion:Have build hBD2 gene eukaryotic expression plasmid pCMV-hBD2 successfully;confirmed the hBD2 have the activity of inhibit the BGC823 cells line proliferation and colony formation,but didn’t effected the GES-1 cells line;the hBD2 also decreased the migration and invasion in the BGC823 cells line;The anti-tumor activities of hBD2 may be according inhibited the expression of MMP2. |
