In order to achieve the purpose of reflecting the appropriate number of chromatographic peak and the known components on the chromatogram different solvents, extraction methods were used to process Bu-Zhong-Yi-Qi Wan (BZYQW), various chromatographic conditions were established to optimization. We aim to explore the fingerprint and determine the main compounds through a sample processing, a establishment of the chromatographic condition. Pharmacodynamics research about tonifying qi and enriching blood of BZYQW were done, use statistical software to establish spectrum- effect relationship. BZYQW originated from the famous Bu-zhong-yi-qi-Tang, which is mainly used for the treatment of spleen descending, qi descending and immune related diseases. The occurrence of tumor and the immune system were closely related, so we studied the inhibitory effect of BZYQW to tumor cells.There are five parts in this dissertation:Chapter one: The chromatography fingerprint study of BZYQW Different solvents, extraction methods were used to process BZYQW, various chromatographic conditions were established to optimization, in order to obtain maximum peak capacity to establish HPLC-DAD and HPLC-ELSD fingerprint. Analysis the fingerprint of 10 batches BZYQW, use traditional Chinese medicine (TCM) fingerprint computer aided similarity evaluation software to analyze map processing.Chapter two:Spectrum-effect relationship of tonifying qi efficacy of BZYQW Qi deficiency mice model was induced by controlling diet and fatigue test; body weight, viscera index, clearance index and correct clearance index were determined. The spectrum-effect relationship was studied using gray relational grade. Result showed that BZYQW have obvious improvement for qi deficiency symptoms of mice; There is certain corresponding relation between HPLC-DAD, HPLC-ELSD fingerprint and tonifying qi efficacy of BZYQW. Hesperidin, formononetin in BZYQW play big role in tonifying qi.Chapter three:Spectrum-effect relationship of enriching qi efficacy of BZYQW Copy a mouse mode of blood deficiency by subcutaneous injection hydracetin (APH); select hematocrit (HCT) and liver weight index as the pharmacodynamic indicators, study the spectrum-effect relationship using gray relational grade method. Results showed that 10 batches of the BZYQW have good improvement for APH caused blood deficiency symptoms. Their are certain corresponding relation between HPLC fingerprint and the role of the blood, so as HPLC-ELSD fingerprint.Chapter four:Simultaneous determination of main components in 10 batches BZYQW On the basis of the fingerprint, through screening detection wavelength, qualitative identification and quantitative determination were done with the chromatographic peak of higher degree of separation, larger peak area. A method is established by HPLC-DAD-ELSD of simultaneous determination of the 10 main components in the Chinese medicine.Chapter five:The study of anti-tumor function from BZYQW Groping towards S180 tumor-burdened mouse model, set up prevention group and treatment group. By detecting mouse viscera index, the index of macrophage cell in abdominal cavity, the index of spleen cell proliferation, cell factor to evaluate repair of BZYQW on S180 tumor-burdened mice, the results showed that supplement of BZYQW have a certain degree of increase in mice anti-tumor immunity, and have certain prevention and cure function to vaccinated mice.
