The microcirculation carries a lot of physiological and pathological information, which plays an important role in the occurrence and development of diseases. The parameters of the microcirculation are the important reference index for clinical diagnosis of diseases, since the improvement of microcirculation image quality is the key for diagnosing of diseases.In this paper based on the comparative analysis of various imaging techniques, choose orthogonal polarization spectral imaging(OPSI) method to achieve image of microvascular.The main research of this paper includes:1、Analyzed the function of each part and set up OPSI device. The choose of the depth of the tissue imaging is realized by changed the angle between the bias and the detector.2、Proposed a dual wavelength dual camera orthogonally polarized microcirculation imaging device, which is combination orthogonal polarization spectral have high resolution imaging and changing the polarization angle can achieve selection of tissue imaging depth. The choice of light source of two different wavelengths, more useful information can be obtained. The principle and the connection mode of the device are introduced in detail.3、Animal liver in vitro and animal ear and the human body nailfold microvessel were observed using the microcirculation imaging device, and classification for the image.4 、 Using digital image processing technology to realized image enhancement processing and vessel segmentation. This paper realized image enhancement respectively from space domain and frequency domain two aspects, compared with the spatial domain, frequency domain image enhancement method of Gaussian matched filter and multi-scale wavelet transform can effectively suppress the noise is introduced. The threshold segmentation and region growing method was used to segment the target microvascular. The threshold segmentation can realize efficient segmentation of tiny blood vessels, the region growth method have a high requirements for the image quality.5、About the assessment of the microcirculation, nailfold microcirculation weighted integral method,which is proposed by Tian Niu is the most widely used in clinic. But due to the complexity of the process, and need certain professional knowledge, poor portability. Nailfold microcirculation imaging instrument parameter measurement software for certain artificial intervention, due to the existence of human error, have poor reliability and low efficiency of measurement results. Therefore, it is urgent to develop an automatic measurement method. First of all was realize the enhancement and target extraction of microvascular image, extraction center line and the measurement of blood vessel diameter and curvature is realized. The blood vessel skeleton and original images are superposed, a high degree of matching; compared the vascular skeleton and the standard center line of manual extraction, mean and variance were less than two pixels. Realized the measurement of the micro vascular diameter and curvature in ultimately. The parameters can be used for classification and evaluation of microvascular. Compared with manual measurement, the advantages of automatic measurement is fast, accuracy and have a high portability, which is the main research direction of micro vessel parameters measurement.
