Bruise is a common problem after surgery. The traditional surgical sutures can cause infection, tissue necrosis and other issues with complicated operation, which will lead to bruises. In recent years, medical tissue glue is widely used in a variety of surgical procedures, which can alleviate the suffering of patients, but also promote wound healing and repair, reduce complications. In this way, the formation of bruises is effectively reduced. Currently, the prepared materials used in domestic are mostly imported with high cost. Therefore, it is of great significance that prepares a low-cost medical tissue glue to reduce surgical scar.In the paper, the gelatin / carboxymethyl medical tissue glue was prepared by blending gelatin, carboxymethyl chitosan and sericin, then crosslinking reaction in the water-soluble carbodiimide. All of this happened under the condition of water bath. The glue were characterized by infrared spectroscopy and scanning electron microscopy about its microstructure and physical properties. Besides, the tissue glue was evaluated and studied as biomedical materials preliminary, through the blood compatibility test, skin irritation test, sensitization test, acute systemic toxicity test, degradation experiments and cell toxicity tests.IR characterization confirmed that the combination was produced among gelatin,sericin and carboxymethyl chitosan.;The complex group of uniform pore space and are communicated with each other, this tissue cells on the surface of the attachment, growth and proliferation of provide a suitable environment for the growth, also helps promote healing of wound; by adjusting the bonding strength of gelatin / carboxymethyl chitosan ratio, curing time, concentration of cross-linking agent and cross-linking agent content adjustment material, with gelatin / carboxymethyl chitosan ratio of G9C1, curing10 min, concentration of cross-linking agent 10%, crosslinking agent content of1% bonding strength is the highest, G8C1 is the second; in vitro degradation experiment results show: the material can be degraded gradually, and through adjusting the composition ratio regulating degradation rate, in order to meet the application requirements different.Hemocompatibility results showed that: the highest rate of hemolysis 1.55 %, far lessthan 5%, having a certain anti- clotting effect. The max andplatelet consumption ratewas22.56%, lower than 40%, in line with the national standard. Skin irritation results showed that both the experimental and negative control group had no erythema or edema phenomenon, which indicated the material was no irritation to the skin. Sensitization test results showed that: the experimental group had no erythema or edema same as the control,with zero sensitization rate. Cytotoxicity test results showed that, the cytotoxicity ratings of prepared glue at 0 or 1 which was less than 2, indicating that the material is in line with the national standard, compared to commercially available α-biological cyanoacrylate glue with cytotoxicity at 2. Systemic toxicity results found that rats have no any toxic symptoms next to the cage observation, compared with the control group, weight relative growth and viscera coefficient of the experimental group have no significant statistical differences,organs of rats do not appear obvious edema or bleeding in pathology examination.Pathological results showed that various organs of heart lung and kidney liver and spleenmicro graph display normal, does not appear swollen, dark area congestionn, rupture, nuclear dissolution, oozing of blood and other pathological changes, good cell morphology normal, that every organ in the body under the action of medical adhesive in acute rat did not happen pathological lesions, is safe and non-toxic.The results showed that the prepared the gelatin / carboxymethyl medical tissue glue had a good ability of sustained-release, nontoxic and safe and had great significance for solving the suture and bruise.
