Abstract/Summary:
Objective To discuss the anti-proliferation effect of Chinese medicine matrine on human hepatoma cancer HepG2 cells and influence on radiosensitivity; to study the protein expressions of HCCR-1 gene and its related genes by X-ray irradiation combined with traditional Chinese medicine matrine on HepG2 cells.Methods 1.HepG2 cells were cultured in vitro and observed the growth of the cells under light microscope. The cells in the logarithmic phase were chose in the experiment.2. The cells were treated with different concentrations of matrine solution by 0.2 mg/ml,0.4 mg/ml, 0.8 mg/ml,1.6 mg/ml,3.2 mg/ml.3. The cytotoxic inhibitory ratio of matrine on HepG2 cells was tested by MTT assay.4. The clonogenic assay was carried out to explore the effects of radios-ensitivity by Chinese medicine matrine on HepG2 cells.5. Apoptotic morphological changes were investigated with Hoechst33258 fluorescence staining.6.HCCR-1 and related genes protein expression levels were measured by Western blotting after treating with matrine for 24h.Results 1.MTT assay showed that the higher the concentration of matrine was, the more obvious its human hepatoma HepG2 cell proliferation inhibitory effect was. The suppression effect showed in some time-dependent and dose-dependent manner.2.Clonogenic assay showed that matrine pretreatment followed by X-ray irradiation was significantly lower than irradiation alone group(P<0.01),and it indicated matrine had a significantly radiosensitivity effect on the HepG2 cells.3.Hoechst33258 staining results showed that after X-ray treatment 24h the combination group showed more obvious morphological changes of cell apoptosis.4.The Western Blot results showed lower protein expression level of HCCR-1 and higher protein expression level of p53 and Bax in combined group compared with irradiation group (P<0.05).Conclusion 1.Matrine can inhibit the proliferation of HepG2 cells in vitro cultivation and increase their radiosensitivity.2. Matrine can reduce the proto-oncogene HCCR-1 protein expression level of HepG2 cells, at the same time it enhanced p53 and Bax protein expression levels.3.Matrine has a cell proliferation inhibitory role on human hepatoma HepG2 cells.The mechanism may be associated with decreasing HCCR-1 gene expression level, increasing its related genes p53, Bax expression levels,which inhibit HepG2 cell proliferation and promote its apoptosis.
