ObjectiveTo observe the effects of Docosahexaenoic Acid( DHA) on the inflammation response in rat after severe burn,explore its mechanism of inflammatory mediators and the lung protectin.Methods160 SD rats were randomly divided into four groups: sham injury group, sham injury + DHA group, scald group and scald + DHA group. scald group and scald + DHA group were inflicted with 30%TBSA full-thickness scald on the back immersing 95°C water for 18 s, sham injury group and sham injury + DHA group were immersed 20°C room temperature for 18 s to simulate the scald process.Sham injury group and scald group of rats injection of saline in 1 m L/ kg by tail vein, sham injury +DHA group and scald +DHA group of rats injection of DHA in 1ml / kg by tail vein 5 minutes after injury.To observed the TNF-a、IL-6N leukotriene B4(LTB4) levels in the serum at 3,6,12,24,48 h five points after injury both four groups; Western Blot assay to observe the expression of NF-k B in lung tissue. Date were processed with one-way analysis of variance, analysis of variance of factorial design and LSD-/ test.Results:1、 The serum contents of TNF-a of rats in scald group and scald +DHA group were obviously higher than those of sham injury group and sham injury +DHA group(7^4522.235, 尸<0.01).3、6、 12、24、48h after injury, in group scald,TNF-a contents were respectively( 157.9士6.2)、( 268.5士6.9)、( 360.4士 13.2) 、(278.1士8.9) 、( 2 14.2士6.1) pg/m L, and in group scald +DHA were respectively(118.3士3.2) 、( 207.3士3.2) 、( 265.4士 12.3) 、( 210.7士8.4) 、( 166.4士9.2)pg/m L, and they are significantly higher those in group sham injury(70.7士2.1) 、(73.4士 1.4) 、( 86.5士5.1) 、( 82.2士2.3) 、( 78.5士1.1) pg/m L(/values from 11.800 to 40.357,P values below 0.01).Group scald +DHA were obviously lower than group scald(/ values from-8.331 to-17.643, P values below 0.01).2、 The seram contents of IL-6 of rats in scald group and scald +DHA group were obviously higher than those of sham injury group and sham injury +DHA group(F=4005.231, i)<0.01).3N 6n 12 n 24n 48 h after injury, in group scald, IL-6 contents were respectively( 168.7士5.4) 、( 210.2士6.2) 、( 306.8士7.2) 、( 250.1士4.2) 、( 201.9士5.3) pg/m L, and in group scald +DHA were respectively(126.5士1.2) 、(160.2士3.1) 、( 230.5士2.2) 、( 183.1 士6.4) 、( 152.3士2.1) pg/m L, and they are significantly higher those in group sham injury( 80.3士2.3) 、(82.3士1.2) 、(85.7士4.2) 、( 84.2士 1.3) 、( 82.5±2.0) pg/m L(/values from 10.334 to 40.357,P values below 0.01).Group scald +DHA were obviously lower than group scald(/ values from-6.332 to-21.596, lv a lu e s below 0.01)3、 The serum contents of LTB4 of rats in scald group and scald +DHA group were obviously higher than those of sham injury group and sham injury +DHA group(7^4372.496,尸<0.01).3、6、 12、24、48h after injury, in group scald, LTB4 contents were respectively( 2.70士0.23)( 3.69士0.24)( 4.59士0.29)( 3.93士0.12)(2.36士0.22) ng/m L, and in group scald +DHA were respectively( 1.76土0.21)(2.02士0.15)( 2.85士0.32)( 1.97士0.33)( 1.68士0.28) ng/m L, and they are significantly higher those in group sham injury( 0.78士0.11)( 0.79土0.11)(0.83士0.13)( 0.80士0.15)( 0.79士0.20) ng/m L(/values from 7.501 to 38.962,P values below 0.01).Group scald +DHA were obviously lower than group scald(/ values from-2.532 to-19.244, P values below 0.01)4、 The protein expressions of NF-k B in lung tissue of rats in group scald and group scald +DHA were significantly higher than the group sham injury(F=5930.414, ^<0.01). The protein expressions of NF-k B in lung tissue of rats in group scald wererespectively( 2.67士0.21) 、( 3.65士0.15) 、( 4.46士012) 、( 3.88士0.14) 、(2.37士0.21),and the group scald+DHA were respectively( 1.62士0.09) 、(2.12士0.15) 、( 2.94士0.21) 、( 1.95士0.16) 、( 1.56士0.17), and significanthigher those in group sham injury( 0.41 士0.09) 、( 0.42士0.08) 、( 0.45士0.09)(0.42士0.09) 、( 0.45士0.12)( / values from 15.1637 to 98.074,尸 values below0.01) 。 The protein expressions in Group scald +DHA were obviously lower than group scald( t values from-14.415 to-30.833, P values below 0.01)Conclusion:DHA can reduce the serum contents level of TNF-a? IL-6? LTB4 and the expression of NF-k B in lung tissue in the rat after burn...
