1.background and objectiveObesity is regarded as a risk factor of cardiovascular disease. BMP4 plays an important role in pluripotent cells generating adipose cells, which has close connection to the amount of adipose tissue in body. BMP4 can promote inflammation, which leads to disorders of BVE(blood vessel endothelium) and cardiovascular disease. BMP7 also plays an important role in pluripotent cells generating adipose cells, but it can protect artery endothelium because of its characters in anti inflammatory and anti fibrosis.The experiment is studying the expression of BMP4 in obese people and the expression of BMP4, 7 in heart, arteries and adipose tissue of obese mice, analyzing the relationship among BMP4, BMI and arterial vascular inflammation and discussing the relationship between arterial vascular inflammation and ratio changes of serum BMP4/BMP7 in obese mice. 2.Methods2.1 The healthy people BMI>24kg/m2 were the fat group, and the BMI< 24kg/m2 were the normal group. Take blood 5ml, empty stomach and ulnar vein. Measure the expression of BMP4, IL-1β, IL-9, TC, TG, LDL, HDL, FPG and hCRP.2.2 Eight four-week-old ob/ob mice were used as the experimental group and 8 C57BL/6 mice were used as the control group. After 8-week full diet, the blood, abdominal aorta and inguinal adipose tissues were collected after all animals were sacrificed. Examine the expressions of BMP4, BMP7, IL-1βand IL-9 by ELISA. Measure the expressions of BMP4 protein in each organization by immunohistochemical and semi quantitative detection localization. The expressions of protein and mRNA of BMP4, BMP7, IL-1β, IL-9 and p-Smad1 were detected by quantitative PCR and Western blotting. 3.Results3.1 People: The serum expression of BMP4 in the fat group was significantly higher than that in the normal group, which is positively correlated to BMI, hCRP, IL-1βand IL-9.3.2 Mice: The serum expression of BMP4 in the experimental group was significantly higher than that in the control group and no significant changes in BMP7 expression were found between the two groups. Ratio of BMP4/BMP7 in the experimental group was significantly higher than that in the control group(P<0.05). Expressions of BMP4 protein and mRNA level in the arteries and myocardial tissue in experimental group were higher than those in the control group, but they were lower in adipose tissues in experimental group. BMP7 protein and mRNA expressions showed no clear difference in arteries and adipose tissues between the two groups. Ratio of BMP4 mRNA/BMP7 mRNA showed obvious difference in the arteries between the two groups(P<0.05), but no significant difference in adipose tissues was observed between the two groups. Inflammations measured by mRNA and protein expressions of IL-1β and IL-9 in the arteries, Myocardial tissue and obesity in experimental group were higher than those in the control group(P<0.05). 4.Conclusion4.1 The serum BMP4 level is higher in fat people, which is positive related to inflammation.4.2 The expression of BMP4 is higher in myocardial tissue and arteries, which is positive related to inflammation.4.3 The ratio change of BMP4/BMP7 is related to arterial vascular inflammation in obese mice.
