Objective To analyze the relationship between fetuin expression in cumulus cells and in Vitro fertilization-embryo transfer outcomes.So that it can offer us an objective and quantitative criteria for oocyte quality evaluation and embryos selectionSubject and methods1. In Anhui Provincial Hospital Affiliated to Anhui Medical University, a retrospective analysis was performed for 64 infertile patients in the period of January2013 to December 2014. Divided into two groups, experimental group and 30 cycle of low for fertilization The control group on the same day to choice the eggs, but a total of 34 patients with normal fertilization cycle. The reasons for infertility are fallopian tube obstruction, The exclusion criteria were uterine malformations, chromosomal abnormalities and poor ovarian response. Comparison between the two groups of patients in general, and pregnancy outcome。Statistical analysis was performed using SPSS software(version17, USA); compared with the rate of chi-square, mean compared with t- test < 0.5 was considered statistically significant.2,The m RNA and protein expressions of fetuin was detected by real-time PCR and Western-Blot in two kinds of cumulus cells Patients with low fertilization and Patients with normal fertilization. Using immunofluorescence and immunohistochemical staining and determine determine the fetuin expression in cumulus cells.Two groups of patients Fetuin levels in follicular fluid extracts were measured using a ELISA kitResults1. The clinical data of two patients with IVF:no differences were detected the age,BMI, duration of infertility, basic hormone, total of Gn doses, endometrial thickness on HCG day, E2 value on HCG day and the number of cycles between these two groups.(P>0.05) 。 Compared to control group, the rates of fertilization was significantly decreased in group experimental(P<0.05)2. The protein Fetuin was expressed in cumulus cells and it was located in cytoplasma and nucleus by immunofluorescence and immunohistochemical staining.Fetuin m RNA was expressed in cumulus cells by quantitative real time PCR.By real-time PCR and Western-Blot, we detected that the m RNA and protein expression of fetuin were decreased significantly in the low fertilization group compared to the normal fertilization group.Using ELISA kit,we detected that the protein expression of fetuin were decreased significantly in the follicular fluid with low fertilization group compared to the normal fertilization groupConclusions Fetuin and its m RNA are expressed in cumulus cells.To some extent,the expression of fetuin may serve as a predictor of fertilization outcome,the level of fetuin and fetuin m RNA expression are Positive correlation with fertilization...
