| ObjectiveDetected the expression of ERCC1 and TUBB3 in advanced gastric carcinoma and adjacent tissue by immunohistochemical method and analyzed the relationship among them,In order to explore the correlation and clinical significance between themMethodsCollected 66 patient’s clinical data and paraffin blocks of advanced gastric carcinoma and 40 adjacent paraffin blocks, tested expression of ERCC1 and TUBB3 with im-munohistochemical method and analyzed expression, clinical data, relevance and pro-gnosis of patients between of them. All datas were analyzed by SPSS17.0 software.Results(1) Expression of ERCC1:ERCC1 was expressed in advanced gastric carcinoma tissues and the adjacent tissues, the highly expression rate of ERCC1 in advanced gastric carcinoma tissues was59.1%,it was higher than the adjacent tissues 30%, the difference has statistically significant(x2=8.443, P＜0.01). No differences of ERCC1 protein was found among patients with different genders, ages, tumor size, invasion depth, lymph node metastasis and tumor site, but all related histological grade of differentiation(P<0.05)。the highly rate of middle-low differentiation was significantly higher than the mucus, signet ring and undifferentiated carcinoma group (x2 = 10.267, P = 0.014).(2) Expression of TUBB3:TUBB3 was expressed in advanced gastric carcinoma tissues and the adjacent tissues, the highly expression rate of TUBB3 in advanced gastric carcinoma tissues was13.6%,it was lower than the adjacent tissues 72.5%, the difference was statistically significant(x2=37.524, P＜0.01). No difference of TUBB3 protein was found among patients with different genders, ages, tumor size, invasion depth, lymph node metastasis and tumor site, but all related histological grade of differentiation(P＜0.05),the highly rate of middle-low differentiation was significantly lower than the mucus, signet ring and undifferentiated carcinoma group (x2= 4.414, P= 0.036).(3) The Relationship between ERCC1 and TUBB3:In all cases of advanced gastric carcinoma tissues, the low expression of ERCC1 was 57 cases including 20 cases of TUBB3, the high expression of TUBB3 was 9 cases including 2 cases of ERCC1, ERCC1 and TUBB3 protein expression in advanced gastric carcinoma was negatively correlated (r=-0.298,P<0.05)Conclusions(1) ERCC1 and TUBB3 were all expressed in advanced gastric carcinoma and adjacent tissues.(2) ERCC1 and TUBB3 expression in advanced gastric carcinoma was negatively correlated, mutual inhibition each other and participate in occurrence and deve lopment.(3) The expressions of ERCC1 and TUBB3 were statistical significance associated with degree of differentiation, which can be used to judge the malignant degree of advanced gastric cancer and have a role in judging the prognosis.(4) ERCC1 and TUBB3 have different expression in advanced gastric carcinoma, ERCC1 and TUBB3 has relationship with platinum and paclitaxel drug-related, By detecting the expression level in gastric carcinoma, and provide a theoretical basis for individualized treatment of advanced gastric cancer. |
