Compare The Protective Effect Of Periplaneta American And Its Different Extracts On Acute Immune Liver Injury In Mice

Liver is the body’s major metabolic organ, assume a variety of nutrients, drugs, metabolic toxins transformation task, so the factors causing damage to the liver can cause serious damage to the human body. Viruses, alcohol, drugs, parasites, autoimmune, nutritional and metabolic disorders, and other factors can cause acute or chronic damage to the liver, of which the immunologic liver injury is the most common and complex. After the acute phase of autoimmune inflammation, is a recurring cause chronic liver damage, chronic persistent injury makes the gradual loss of normal liver tissue structure, and gradually develop into cirrhosis, liver cancer. Therefore, pathogenesis and drug development for the acute phase of autoimmune liver damage is always the focus of basic and clinical research and hotspots. China has a long history of Chinese medicine treatment of liver disease, looking from the treasure house of Chinese herbal medicine to treat autoimmune liver damage drugs have important clinical implications.Objective To compare the protective effect of Periplaneta American and its different extracts on concanavalin A(Con A)-induced acute immunological liver injury in mice. Preliminary identify the main ingredients of its different extracts, and investigate its effective active sites.Methods The model of mice acute immunological liver injury was induced by Con A on day 10 after pretreatment with Periplaneta American and its different extracts.After 8 h,the mice were weighted,and blood were collected for determinating serum alanine aminot Iansferas(ALT) and aspartate aminotransferas(AST). Then the mice were killed and the liver and spleen were weighted immediately,and the liver and spleen index were calculated. The liver homogenates were prepared to test malondiadehyde(MDA),superoxidedismutas(SOD), glutathione(GSH), tumor necrosis factor-α(TNF-α), interferon-γ(IFN-γ)and hepatic pathological examination was observed with HE coloration.Results Periplaneta American and its different extracts might have the composition of Amino acids, protein polypeptide, sugar and organic acids. Compared with model group, Periplaneta American and its different extracts could decrease liver and spleen index( P<0.05, P<0.01) and the activity of ALT,AST( P<0.05, P<0.01) in serum,the content of MDA( P<0.05,P<0. 01), TNF-α( P<0.05, P<0.01) and IFN-γ( P<0.05, P<0.01) in liver. Moreover, it could enhance the SOD and GSH activity( P<0.05, P<0.01) in liver tissues and thus relieved liver pathological injury. The efficacy of Periplaneta American and its different extracts existed differences.Conclusion Periplaneta American and its extracts have protective effects on acute immunological liver injury in mice,and the effect of protozoa and water extracts is better than alcohol extracts and alcohol-water double extracts.
