Objective To detect the expression of miR-200s (miR-200a, miR-200b, miR-200c, miR-141 and miR-429), ZEB1 and E-cadherin in nasopharyngeal inflammation tissues and Nasopharyngeal carcinoma (NPC) tissues. To investigate the correlation of miR-200s, ZEB1 and E-cadherin expression with clinicopathological parameter of NPC and the interrelationship between them, in order to evaluate the effect of them in the invasion and metastasis of NPC.Methods Expression of miR-200s, ZEB1 mRNA and E-cadherin mRNA in 12 cases of nasopharyngeal inflammation and 39 NPC tissues were detected by using Real-time PCR method. Studied the interrelationship between them and the correlation of them with clinicopathological parameter(gender, age, T stage, N stage and clinical stage) of NPC. Immunohistochemical staining was performed to detect the expression of ZEBland E-cadherin in NPC tissue, analyzed the association between them and the relationship of them with clinicopathological parameterof NPC.Results 1.The expression of miR-200a,miR-200b and miR-200c were lower in NPC tissues than that in inflammation tissues, miR-200a and miR-200b decreased significantly (p<0.05). Expression of miR-141 and miR-429 were higher in NPC tissues than that in inflammation tissues (p>0.05). Abnormal expression of both miR-200a and miR-200b in NPC were significantly associated to T stage, N stage and clinic stage (p<0.05), but the difference of their expression between the different gender, age were not statistically significant (p>0.05). The different expression of miR-141, miR-429 and miR-200c in NPC between the different gender, age, T stage, N stage and clinic stage were not statistically significant (p>0.05)2. Expression of zebl mRNA was higher in NPC tissues than that in inflammation tissues (p<0.05), but the expression of E-cadherin mRNA was lower in NPC (p<0.05). Abnormal expression of zeb1 mRNA in NPC were significantly associated to T stage, N stage and clinic stage (p<0.05), but the different expression between the different gender, age were not statistically significant (p>0.05). Expression of E-cadherin mRNA in NPC were significantly related to N stage (p<0.05), but the different expression between the different gender, age, T stage, N stage and clinic stage were not statistically significant (p>0.05)3. Expression of miR-200a was negative correlated with the expression of ZEB1 mRNA(r=-0.372,p=0.020), the expression of ZEB1 mRNA was negative correlated with the expression of E-cadherin mRNA (r=-0.349, p=0.03); the differences were both statistically significant (p<0.05)4.The positive expression frequency of ZEB1 protein in the NPC tissues was 71.8% was significantly higher than that in inflammation tissues (p<0.05). The expression of ZEB1 protein in NPC were significantly associated to T stage (p=0.017), N stage (p=0.004) and clinic stage (p=0.017), but the different expression between the different gender, age were not statistically significant (p>0.05)The positive expression frequency of E-cadherin protein in the NPC and inflammation tissues was 20.5% and 91.7% respectively, the difference was statistically significant (p<0.05). The expression of E-cadherin protein in NPC were significantly related to N stage (p=0.016), but the different expression between the different gender, age, T stage, N stage and clinic stage were not statistically significant (p>0.05)5. In NPC tissues, the expression of ZEB1 protein was negative correlated with the expression of E-cadherin protein (r=-0.387,p=0.015)Conclusion 1. miR-200a and miR-200b may play a role of cancer suppressor gene in NPC. miR-200a may control the expression of ZEB1 and weaken its inhibition of E-cadherin, thereby inhibiting the invasion and metastasis of NPC.2. ZEB1 may promote the invasion and metastasis of NPC by inhibiting the expression of E-cadherin.3. Abnormal expression of miR-200a, miR-200b, ZEB1 and E-cadherin may be associated with invasion and metastasis of NPC, but their definite mechanism needs further research.
