objective:Osteoporosis (Osteoporosis, OP) is a systemic disease due to an imbalance between osteoblast and osteoclast in bone remodeling, decrease of the quantity and activity of osteoblast, while increase of the osteoclast, which leds to resorption much stronger than bone formation, finally, resulting in osteoporosis. Numerous studies demonstrate that adipose-derived stem cells with osteogenic potential. Influence of osteoporosis on the biological character and osteogenic differentiation of adipose derived stem cells have not been completely stated. This experiment aims to explore the influence of osteoporosis on osteogenic and differentiation of adipose derived stem cells by comparing the osteogenic ability of adipose-derived stem cells derived from the osteoporosis mice and the normal, which lays the foundation for autogenous adipose-derived stem cells transplantation and bone defect repair for osteoporosis patients.Methods:Female C57BL/6 mouse were randomly divided into experimental group (OVX group) and control group (sham operation group). Adipose derived stem cells were obtained from inguinal adipose tissue of two groups, which were purified and amplified in vitro to the third generation. Osteogenic ability and difference between two groups were tested using alizarin red staining after induced and differentiated adipose derived stem cells into osteoblasts; MRNA of osteogenic related gene and the expression of bone related protein of the adipose derived stem cells were detected by real-time PCR, immune fluorescence staining and Western blot.Results:Alizarin red staining showed that calcium deposition of the ASCS source of osteoporosis was significantly less than the control group, while real-time PCR and immunofluo rescence staining showed that, compared with elated gene and the expression of bone related protein of the adipose derived stem cells were significantly decreased in the experimental group. The above results showed that, the osteogenic differentiation ability of adipose derived stem cells came from OVX group were deteriorated.Conclusion:1.Through the removal of both ovaries of female mice, successfully constructed an animal model of osteoporosis;2.The proliferative capacity of OP-ASCs from OVX group significantly decreased than ASCs from sham operation group;3.The osteogenic potential of OP-ASCs significantly decreased than normal.
