Ojective: To investigate the effect of curcumin on brain glucose metabolism in a rat model of intracerebral hemorrhage(ICH), which compaired with the changes of interleukin-6(IL-6) 、 interleukin-1β(IL-1β)、tumor necrosis factor alpha(TNF-α) expressions in rats with ICH and the neurological severity scores. To evaluated the therapeutic effect of curcumin and provided scientific experimental dates for the clinical application of curcumin on ICH. Methods : 48 healthy adult male rats were randomly divided into 4 groups: nomal group(group A), ICH+curcumin group(group B), ICH +Vehicle group(group C), and sham operated group(group D), 12 rats in each group. Acoording to the Rosenberg GA, animal ICH model was made by injecting collagenase Ⅳ1μl into the right caudate nucleus of rats. In ICH+curcumin group, rats received intraperitoneal injection of curcumin immediately when ICH model was built successful. In ICH +Vehicle group: rats received the same amount of vehicle.The sham operated group was made by injecting vehicle 1 μl into the right caudate nucleus of rats, with no treatment. The nomal group with no operating. The 4 groups(group A, group B, group C, group D)were randomly divided into 2 groups respectively(group A and A′, group B and B′, group C and C′, group D and D′), 6 rats in each group. Ingroup A′, group B′, group C′, group D′, every rat was injected 18F-FDG17.8±0.4MBq into caudal vein at 6h、24h、48h、3d、5d、7d、14d after the model built successful, respectively, following anesthesia. Thirty min later, 18F-fluorodeoxyglucose(FDG) micro-PET/CT scans were acquired at the 8 time points. According to the micro-PET/CT film, the SUV in the perihematomal brain tissue of ICH was measured and analyzed. In group A, group B, group C, group D, every rat was withdrew blood serum from the tail vien for IL-6、IL-1β、TNF-α. All dater used one-way analysis of variance when they are normal distribution, and Student ’ s t test was used to identify the changes of SUV and the levels of IL-6、IL-1β、TNF-αbetween group B and group C, group B and the mirror side, group B and group D, respectively. The changes of SUV is significant when the P<0.05. Results: 1. At the acute stage of ICH the neurological severityscores in group B was lower significantly than group C, and higher than group D.2. Micro-PET/CT clearly showed the normal brain structure and ICH structure of glucose metabolism on rats. After cerebral hemorrhage, the hematomal and perihematoma brain tissue was obviously decreased on glucose uptake. 3. ① In group B: After 6h, the 18F-FDG density in perihematomal brain tissue of ICH decreased(SUV 2.70±0.19)and was lowered than the mirror side( SUV 3.10±0.22), which is statistical significant(t=2.594, P=0.023,P<0.05);the 18F-FDG density reached the minimum at 24h(SUV 1.20±0.08), began to increase at the next time points. The changes of SUV is significant in the group B( t=9.084,P=0.0001, P<0.05). ② Each time point of glucose metabolism in group B was lower significantly than group D(t=7.693,P=0.0001,P<0.05), and higher than group C(t=2.214,P=0.047,P<0.05). 4. In group B: After 6 h,the levels of IL-6、 IL-1β、 TNF-α in blood serum increased and was peaked at 3 d, and decreased at the next time points. Each time point of the levels of IL-6、IL-1β、TNF-α in group B was lower significantly than group C, and higher than group D. Conclusion:1. micro-PET/CT clearly showed the normal brain structure and ICH structure of glucose metabolism on rats. After cerebral hemorrhage, the hematomal and perihematoma brain tissue was obviously decreased on glucose uptake.2.Curcumin owned neuroprotection effect on ICH, as it could effectively improve the glucose uptake on hematomal and perihematoma brain tissue and neurological severity scores, reduce the levels of IL-6、IL-1β、TNF-αafter ICH.
