Abstract/Summary:
Objective To investigate the effect of NBS1 knockdown by RNAi combined with cisplatin on proliferation and apoptosis ability of human triple negative breast cancer MDA-MB-231 cells.Methods The NBS1 was indentified overexpression in breast cancer tissue by IHC. The MDA-MB-231 cells were infected by the adenovirus vector with NBS1-siRNA plasmid containing. The downregulated expression of NBS1 gene was determined by Western blotting. Changes of cell proliferation under 15μg/ml cisplatin were detected by CCK-8 assay. The changes of apoptosis and cell cycle were analyzed by using FCM.Results The result of Western blotting confirmed NBS1 gene down regulated by RNAi. The average optical density of NBS1-siRNA group was lower than negative and blank control groups by CCK-8 assay(P<0.05). The apoptosis rate of NBS1-siRNA group was higher than that of negative and blank control groups evaluated by FCM.Conclusion Knockdown of NBS1 gene suppressed the proliferation and promoted apoptosis of human triple negative breast cancer MDA-MB-231 cells. Cisplatin combined with NBS1-siRNA was obviously more effectively cytotoxic than cisplatin alone in human triple negative breast cancer MDA-MB-231 cells.
