Objective To investigate the association between single nucleotide polymorphisms (SNPs) in FIFO-ATPase genes and HGF gene in high risk families for liver cancer in Fusui Country,GuangXi.Methods In case-control study,21 liver cancer family groups were selected as cases and 10 control family groups.PCR-RFLP were adopted to cpmpare and analyse their F1FO-ATPase genetic polymorphisms rs2298787、 rs1264896、rs2291535 and HGF gene polymorphism rs5745652.Then logistic regression model were used to analyse the relationship between F1F0-ATPase and HGF genetic polymorphisms of the above sites and susceptibility to the heredity of hepatocellular carcinoma(HCC).Results Genotype of rs2298787、rs1264896、rs2291535、rs5745652 in liver cancer family groups were consistent with Hward-weinberg equilibrium.The samples detected were representative and comparable.1、There were CC,AC and AA three genotypes in rs2298787C/A, the difference of the frequence distribution between the two groups was statistically significant (X2=8.034 P=0.018).Comparing with CC genotype, AC genotype was associate with susceptibility to hepatocellular carcinoma(P=0.001, OR=0.663,95%CI=0.41-0.92). AA genotype was associate with susceptibility to hepatocellular carcinoma (P=0.005, OR=0.702,,95%CI=0.49-0.81).2. There were CC,TC and TT three genotypes in rs1264896C/T, the difference of the frequence distribution between the two groups was not statistically significant (X2=1.094 P=0.296).Comparing with CC genotype TC genotype was not associate with susceptibility to hepatocellular carcinoma (P=0.057 OR=2.013,95%CI=0.78-5.31). TT genotype was not associate with susceptibility to hepatocellular carcinoma (P=0.281, OR=2.234, 95%CI=0.31-4.24)3、There were AA,AG and GG three genotypes in rs229235A/G, the difference of the frequence distribution betweenthe two groups was not statistically significant( X2=2.537 P=0.281).Comparing with AA,genotype AG genotype not was associate with susceptibility to hepatocellular carcinoma (P=0.073 OR=3.122,95%CI=0.73-5.56). GG genotype was not associate with susceptibility to hepatocellular carcinoma (P=0.286, OR=2.13, 95%CI=0.91-5.53)4、Genotype of HGF rs5745652 in liver cancer family groups were consistent with Hward-weinberg equilibrium the samples detected were representative and comparable. There were CC,CT andTT three genotypes in rs5745652, the difference of the frequence distribution between the two groups was statistically significant (X2=36.89 P=0.001).Comparing with CC genotype, CTgenotype was associate with susceptibility to hepatocellular carcinoma(P=0.011, OR=17.42, 95%CI=6.5-80.2). TT genotype was associate with susceptibility to hepatocellular carcinoma (P=0.005, OR=1.87,95%CI=1.21-2.32)Conclusions F1F0-ATP synthase gene rs2298787 and HGF gene rs5745652 polymorphisms correlated with liver cancer susceptibility in Fusui Country of Guangxi. Rs 1264896 and rs2291535 polymorphisms was not associate with susceptibility to hepatocellular carcinoma.Risk of HCC is increased in carriers of the rs2298787 C allele and rs5745652 T allele.
