Objective:To investigate the effect of different routes of curcumin on wound healing and wound inflammatory mediators and its mechanism.Methods:40 sterile male C57 mice were randomly divided into four groups, control group (group A), skin Group (Group B), Intraperitoneal administration group (Group C) and the skin and the abdominal cavity combined group (Group D), each group has 10 mice.The skin wound mice model of group A, B, C and D were operated on the back of the mouse by perforated foil.Group A is the control with back wound and intraperitoneal injected the DMSO. Group B is injected the Curcumin into back wound and DMSO into intraperitoneal. Group C is injected the DMSO into back wound and Curcumin into intraperitoneal. Group D is injected the Curcumin into back wound and intraperitoneal in the same time.Each group is injected once a day and last totally seven days. The total amount of curcumin for group B, C and D was given daily lOOmg/Kg. The wound back of each group are taken photos in dayl, day3, day5, day7 respectively.The specimens of back skin around the wound were collected on the third day.The Real-time PCR was used to detect the expression of Interleukin-6(IL-6), interleukin-8 (IL-8) and tumor necrosis factor-α(TNF--α). The growth of granulation tissue, capillary change and inflammatory cell infiltration in skin wound edge were observed on HIM.Results:The group had no difference between the three groups compared with group A, on the third day when the B, C, D tissue wound healing is significantly faster, a group of mice healing rate was 36.7%+2.4%, group B was 52.2%+1.2%, group C was 58.9%+2.3%, group D was 52.8%+1.6%, the difference was significant (P< 0.05), B, C and D were statistically significant (P>0.05). On the 7th day of a mouse wound healing rate was 63.1%+2.2%, group B was 80.4%+1.6%, group C was 77.8%+2.4%, group D was 81.4%+ 1.7% in group A and B, C, D group also significant difference (P>0.05), B, C, D group, the difference was not statistically significant (P<0.05). On the third day B, C, D group a limbal tissue inflammatory cytokines IL-6, IL-8, TNF alpha expression amount compared with the a group decreased significantly (P<0.05), a group of 69.9+2.3, group B 31.1+2.1, group C (32.9+1.2, group D 30.27+ 0.2, B, C and D between the three groups no significant difference (P>0.05).Conclusion:The different routes of curcumin could reduce the inflammatory reaction around the wound and promote the wound healing and have no effect on the route of administration.
