Objective:Explorethe influence of applying Plate-let rich plasma to foot ulcer healing of diabetic rats, Provide the experimental evidence for Plate-let rich plasma applied in the treatment of diabetic food。Methods:Cutaneous ulcer model was produced at 8 weeks after the onset of diabetic rat, Randomly divided into treatment group and control group, ulcers were treated with platelet-rich plasma or N.S. dressing Daily dressing and measurement of ulcer area. The Blood and wound tissues werecollected at 5d and 14d. we examine the levels of MMP-2,MMP-9,TIMP-1 and AGEs with Elisa. The wound tissues were sliced to pathology and observed pathological changes of HE staining.Results:5 days after ucler model seted, the ulcer surface area between PRP treatment group and control group had no difference. The levels of MMP-2, MMP-9 in tissue in PRP treatment group tissue were lower than control group, while the level of TIMP-1 were higher than control group(P< 0.05). The numbers of blood vessels in newborn were more than control group with HE staining.14 days after injury, the ulcer surface area of PRP treatment group was smaller than control group, the difference between the levels of MMP-2, MMP-9 and TIMP-1 in 2 groups was more obvious than before (P< 0.01). The number of fibroblasts of PRP treatment group were more than control group, while infiltration of inflammatory cell was lower than control group. The level of AGEs and the levels of MMP-2, MMP-9, TIMP-1 in blood between 2 groups had no difference at each period.Conclusions:1.The establishment of Diabetic foot ulcers model through burning means was successed2.Platelet-rich plasma treatmentmay through reducing the content of MMP-2, MMP-9 and TIMP-1 improve, stabilize MMP-9/TIMP-1 ratio, accelerate diabetic rats foot ulcer healing with hyperglycemia level.
