Objective:It was to observe the relationship between the two in epith-elial ovarian cancer and its clinical significance though the expression of determination of HMGB1 and MMP-7 in ovarian epithelial tumors.Methods:1 Specimen collection: Third Hospital of Hebei Medical University,Second Affiliated Hospital of northern Hebei medical college 18 cases of normal ovarian tissue, 18 cases of benign epithelial ovarian tumors and 38 cases of malignant epithelial ovarian tumors. HE staining of specimens was for histological diagnosis.2 Using immunohistochemical method of PV to test the expression of HMGB1 and MMP-7 in epithelial ovarian tumors(benign epithelial ovarian tumor tissues 18 cases and epithelial ovarian cancer 38 cases) and 18 cases of normal ovarian tissue.3 Statistical analysis: Data were analyzed by SPSS17.0 software,compared with the expression of HMGB1 and MMP-7 in epithelial ovarian tumors(benign epithelial ovarian tumor tissues, epithelial ovarian cancer) and normal ovarian tissue of two independent samples χ2 test,(P<0.05) indicates that there are statistically significant differences. The correlation between HMGB1 and MMP-7 using Pearson product-moment correlation,(P<0.05)shows the two are correlation.Results:1 Expression of HMGB1 protein in epithelial ovarian carcinoma,benign epithelial ovarian tumor tissue and normal ovarian tissue. It was positive expression that there were brown granules site of the nucleus in normal ovarian tissue, benign epithelial ovarian tumor tissue, epithelial ovarian cancer tissue expression. In this experiment, the positive expression rate of HMGB1 protein expression in the three respectively 78.9%, 16.7% and 5.6%. The difference was significant(P<0.05), comparing the positive expression rate of epithelial ovarian cancer to normal ovarian tissue and benign epithelial ovarian tumor tissue(χ2=34.7, P<0.05). The difference was significant,comparing the positive expression rate of epithelial ovarian cancer to benign epithelial ovarian tumor tissues(χ2=19.58, P<0.05). The difference was significant, comparing the positive expression rate of epithelial ovarian cancer to normal ovarian tissues(χ2=26.62, P<0.05). The difference was no significant, comparing the positive expression rate of benign epithelial ovarian tumor tissue to normal ovarian tissue(P=0.603, P>0.05).(Table 1)2 In 38 cases of epithelial ovarian cancer, it was 46.2%, 92.0%respectively that the expression rate of HMGB1 protein of clinical stageⅠ~Ⅱ and clinical stage Ⅲ~Ⅳ, the difference was statistically significant(P=0.013, P<0.05). It was 68.7%, 100% respectively that the expression rate of HMGB1 protein of G1~G2 and G3, the difference was statistically significant(P=0.009, P<0.05). It was 100%, 66.7% respectively that the expression rate of HMGB1 protein with lymph node metastasis and without lymph node metastasis, the difference was statistically significant(P=0.006,P<0.05). The results showed that the higher the degree of malignancy organization, the higher positive expression level of HMGB1 protein,histological grade, lymph node metastasis and clinical stage affect the degree of expression of HMGB1 protein in epithelial ovarian cancer.(Table 2)3 Expression of MMP-7 protein in epithelial ovarian carcinoma, benign epithelial ovarian tumor tissues and normal ovarian tissue. It was positive expression that there were brown granules site of the cytoplasm and a small amount of cell membrane in normal ovarian tissue, benign epithelial ovarian tumor tissues, epithelial ovarian cancer tissue expression. In this experiment,the positive expression rates of MMP-7 protein expression in the three groups were 73.7 %, 16.7%, 11.1% respectively. The difference was significant,comparing the positive expression rate of epithelial ovarian cancer to normal ovarian tissue and benign epithelial ovarian tumor tissues(χ2=26.86, P<0.05).The difference was significant, comparing the positive expression rate of epithelial ovarian cancer to benign epithelial ovarian tumor tissue(χ2=16.07,P<0.05). The difference was significant, comparing the positive expression rate of epithelial ovarian cancer to normal ovarian tissue(χ2=19.23, P<0.05).The difference was no significant, comparing the positive expression rate of benign epithelial ovarian tumor tissue to normal ovarian tissue(P=1.0,P>0.05).(Table 3)4 In 38 cases of epithelial ovarian cancer, it was 53.8%, 88% respectively that the expression rate of MMP-7 protein of clinical stage Ⅰ~Ⅱ and clinical stage Ⅲ~Ⅳ, the difference was statistically significant(P=0.04, P<0.05). It was 56.2%, 95.4% respectively that the expression rate of MMP-7 protein of G1~G2 and G3, the difference was significant(P=0.005, P<0.05). It was91.3%, 60% respectively that the expression rate of MMP-7 protein with lymph node metastasis and without lymph node metastasis, the difference was statistically significant(P=0.039, P<0.05). The results showed that the higher the degree of malignancy organization, the higher positive expression level of MMP-7 protein, histological grade, lymph node metastasis and clinical stage affect the degree of expression of MMP-7 protein in epithelial ovarian cancer.(Table 4)5 The expression level of HMGB1 protein in epithelial ovarian cancer was positively correlated with the MMP-7 protein(r=0.563, P=0.001).(Table5)Conclusions:1 There were negative or weakly positive expression of HMGB1 and MMP-7 protein in normal ovarian tissue, there were moderate positive expression of HMGB1 and MMP-7 protein in benign ovarian epithelial tissue,there were strong positive and moderate positive expression of HMGB1 and MMP-7 protein in epithelial ovarian tumors, they may be involved in the occurrence of epithelial ovarian cancer.2 HMGB1 and MMP-7 were over-expressed in epithelial ovarian tumor,they were closely related with the histological grade, lymph node metastasisand clinical stage. AS higher the degree malignancy of the ovarian tissue was,the expression of the HMGB1 and MMP-7 protein was higher. They may be involved in the development and metastasis of epithelial ovarian cancer.3 The over-expression of HMGB1 and MMP-7 maybe played an important role in the occurrence and development of epithelial ovarian cancer by interaction.
