T cell is one of the most important cells in our immune system. Targeting to regulate its function can be used for the treatment of a variety of immune-related diseases. But due to the distribution and structure characteristics of T cell, Drug delivery system targeting to the T cell is very difficult to get and even one of the major problems in drug delivery areas.HIV can specifically recognize and combine with CD4+ T cell after going into the human body, then damage the human’s immune system and make people easily get many kinds of disease, inducing the production of AIDS. AIDS started from Africa, then developed into one of the most serious epidemic diseases around the world.HIV can specifically invade CD4+T cell because of that their envelope protein gp120 can specifically recognize the CD4 receptor on the surface of the cell, then the virus envelope fuse with cell and put its core particle into the cell. This mechanism of virus going into the cell becomes the designing origin for a variety of AIDS drugs, then the entry inhibitors arises. But it only can inhibit the spread of virus, and cannot cleared them. The sustainability high cost of medication give them huge pressure on money and the evolved mechanism will also make them fail. But this invading mechanism provides a new way for drug delivery carriers’ designing. At the same time, this carrier can be not only used in the treat of HIV, but also other diseases corelated with CD4+ T cell.Microvesicle(MV) is found recently as a carrier for information communication between cells. It is budding directly from a cytoplasmic membrane, so it has the same structure and composition as the cell membrane. According this feature, it can go into the target cells in the form of membrane fusion. At the same time, because of their coming from the body itself, they will not be identified and cleared by the organism clearing system, this is the external advantage as natural drug carrier.We design one kind of 293T cell which can express steadily gp160, then they can secrete MV containing gp120 and gp41 in the membrane. By the recognising between gp120 and CD4, the drugs packaged in the MV can target to the CD4+T cells,which is the targeting cell of HIV. Our extraneous experiment proved that MV containing gp120 than MV from ordinary 293T is more efficient on targeting to the CD4+ T cell. This proved that it perhaps can be a potential drug delivery carrier targeting to T cell.
