Purpose: To explore the effect of Cdc25 C on radiotherapy sensitivity in human esophageal squamous carcinoma EC9706 cells.Methods: 1) Construct expressing sh RNA against human CDC25 C was generated by subcloning into p GV248, then lentiviral particles were packaged and transfected into EC9706 cells. Stable knockdown of CDC25 C EC9706 cell lines(Knockdown)and stable transfected with empty vector plasmid cell lines(Control) were established.2) The knockdown efficiency was evaluated by detection of protein level with western blot or m RNA level with RT-PCR. 3) The proliferation and radiosensitivity of the two cells treated with various dose of X-rays was assayed by MTT and cell colony formation assay.Results: 1) Stable knockdown of CDC25 C in EC9706 cell lines(Knockdown) and stable transfected with empty vector plasmid cell lines(Control) were established. 2)The proliferation of Control and Knockdown cells was suppressed by treated with increasing dose of X-rays. Furthermore, the inhibiton of proliferation was more obvious in Control cells(p<0.05). 3) Cell colony formation assay found that compared with knockdown group(Knockdown), the survival fraction of Control was decreased more significantly(p<0.05) after treated with various doses of X-ray.Conclusions: Downregulated the expression of CDC25 C may reduce the radiosensitivity and proliferation activity of esophageal squamous cell carcinoma.
