Background and Objective:Endometrial carcinoma is a kind of endometrial epithelial malignant tumor, occurs in the endometrium,of which the adenocarcinoma derived from endometrial glands is the most common. At present,the cure rate in early endometrial cancer is obvious higher than late stages especially with local recurrence or distant metastasis.But the early diagnosis rate of endometrial carcinoma is much lower than that of cervical cancer,Because of the limitation of sampling or misdiagnosed as irregular menstruation.Therefore, It is essential to detect new indexes and explore the mechanism of Endometrial carcinoma’s occurrence, development, infiltration and metastasis,which will have a great significance in clinical diagnosis, evaluation of prognosis of endometrial carcinoma.Inhibition of growth factor family is an important group of tumor growth inhibition factor which is founded in recent years, associated with the occurrence of multiple systems in many tumors.Recent studies have found that, ING4 gene is miss expressed in a variety of tumor tissues, while no one has conducted this study in endometrial cancer.Our purpose is to measure the expression of ING4 gene in Different pathology of endometrial tissue and to investigate its clinical significance, as well as the relationship between its expression and devdopment and prognosis of endometrial carcinoma.Methods:ING4 gene was measured by real-time quantitative reverse transcriptionpolymerase chain reaction(RT-PCR)and immunohistechemical technique(S-P method) in 25 cases of proliferate endometrial and 15 cases of atypical hyperplasia and 45 cases of endometrioid adenocarcinoma.Results:The positive-expression rate of ING4 in endometrioid adenocarcinoma was lower than that in proliferate endometrial and atypical hyperplasia.There were statistic differences among three groups(P<0.05).The RT-PCR result was consistent with the immunohistochemical staining. The expression of ING4 gene m RNA was down-regulated in primary endometrioid adenocarcinoma patients compared the matched tissues.Statistical analysis revealed that the difference between expression of cancerous tissues and the matched tissues was significant(P<0.05). Furthermore,the expression rate of ING4 in endometrioid adenocarcinoma was associated with histologic grade(P<0.05). But it was not associated with the depth of symmetrical tumor invasion and lymphonode metastasis.There were statistic differences(P>0.05). But further studies are needed to indicate the relationship between its expression and pathological stagingConclusion:ING4 gene silencing in endometrioid adenocarcinoma and atypical hyperplasia. The lower levels of ING4 can promotes endometrioid adenocarcinoma cell growth.ING4 may be as one of the significance indicatory in predicting the risk of carcinogenesis from precanccrouslesions.
