Background and ObjectiveThe glaucomas are a group of diseases traditionally defined as having opticnerve damage and intraocular pressure levels that exceed the tolerance of the affected eye. In the aspectof the pathogenesis, academics did not reach the same conclusion, many studies have shown [4-6] the elevated intraocular pressure may cause increased vitreous glutamate concentration in primary open-angle glaucoma,resulting in the ganglion cell apoptosis.Caspase-3 is the key to cell apoptosis protease cascade,the expression levels of Caspase-3 in tissue can reflect the apoptosis stateof the tissue to some extent. Benzothiazole drugs riluzole is a kind of anti glutamic acid drugs, and many scholars abroad explore themechanism of riluzole, pharmacological action and in anesthesia, nerve protection, anticonvulsant, antidepressant, analgesic and anti dependence etc from the whole, in vitro, the molecular level. It is the earliest glutamate antagonists widely used for the treatment of patients with amyotrophic lateral sclerosis, later studies have found that riluzole have stable intracellular ions(sodium, potassium, calcium) concentration effect. Riluzole as glutamate release inhibitors, was ued in Amyotrophic lateral sclerosis treatment, Riluzole has been reported abroad that it can shorten acute ocular hypertension rat retina b-wave recovery time [2]; domestic studies has already carried out the treatment of retinal microvascular abnormalities hyperplasia[3].In this study,bying prepared acute intraocular hypertension rat models with anterior chamber perfusion.Using Riluzole to understand the effect of Riluzole in acute intraocular hypertension of retinal injury,and provide the basis for the treatment of acute intraocular hypertension. Materials and methods80 cases of acute intraocular hypertension rat models was prepared with anterior chamber perfusion.40 cases of high intraocular pressure group、40 cases of treatment group and 40 cases of normal group.The animals from each group were sacrificed at different time-points(12,24,48 and 72 ours) and the eyeballs were removed for HE staining,immunohistochemistry evaluation and TUNEL staining to investigate morphological and molecular changes occuring in the retina.Using SPSS17.0 software for data processing, the results were used x±s,he difference was statistically significant with P <0.05. Results1.HE staining:The normal rats have clear hierarchy on retinal.Result for high intraocular pressure group,the retinal’s cells were edema and disordered.The edema of retina was reduced in treatment group than high intraocular pressure group.2.Immunohistochemistry2.1 Normal group:The expression of Caspase-3 in rat retina was weakly.At different time points,the number of positive cells of Caspase-3 waved at at the same level,it didn’ t have statistical difference.2.2 High intraocular pressure group and treatment group:The expression of Caspase-3 changed with different time intervals after reperfusion,increased first and decreased afterwards.The number of positive cells of Caspase-3 was the most in 24 h.Results there was a statistical difference at different time points.2.3 Effect analysis of Riluzole intervention:The one-way analysis of variance showed that the results of positive cells of Caspase-3 at different time-points were F12h=67.851 P<0.01、F24h=112.187 P<0.01、F48h=39.640 P<0.01、F72h=15.033 P<0.01.There was a significant difference between high intraocular pressure group and treatment group by using LSD-t test.3.TUNEL staining2.1 Normal group:The number of apoptotic cells in rat retina was uncommon.At different time points,the number of apoptotic cells waved at at the same level,it didn’ t have statistical difference.2.2 High intraocular pressure group and treatment group:The number of apoptotic cells changed with different time intervals after reperfusion,increased first and decreased afterwards.The number of apoptotic cells was the most in 24 h.Results there was a statistical difference at different time points.2.3 Effect analysis of Riluzole intervention:The one-way analysis of variance showed that the results of the number of apoptotic cells at different time-points were F12h=93.888 P<0.01、F24h=396.437 P<0.01、F48h=482.622 P<0.01、F72h=198.285 P<0.01.There was a significant difference between high intraocular pressure group and treatment group by using LSD-t test. Conclusions1.The expression of Caspase-3 increased in rat retina with acute intraocular hypertension.Using Riluzole could effectively decrease the expression of Caspase-3.2. Riluzole could reduce the cell apoptosis with acute intraocular hypertension on rat retina,it may have certain protective effect on retina.The mechanisms may be related to Riluzole could inhibit concentration of glutamate.
