ObjectiveTo explore effect of Valproic Acid on Oct-4 in Rat bone marrow mesenchymal stem cells into neuron like cells during differentiation.MethodsThe BMSCs were separated from rat’s bone marrow and cultured in vitro. Experiment groups were divided into experimental group, control group.(1)control group: without VPA, added with pre-induction(mediumcontaining 1μmol/L β-ME,10% FBS in DMEM/F12), 24 hours later, the cells were induced into neurons-like cells by DMEM with 2%DMSO and 200μmol/BHA,and harvested 5h later. experimental group: with the same induction method but receiving VPA. Morphological changes were observed by light microscope,NSE、GFAP、Oct-4 were detected by immunocytochemistry,NSE、GFAP、Oct-4were tested by Western blot.ResultsWith the observation of light microscope,spindle shaped BMSCs after induction showed neuron-like cells shape with long projections,which were different from experiment group with the length and quantity of the projections shorter and less. Western blotting showed that NSE、GFAP of experimental group were significantly higher than those of the control group.ConclusionRat bone marrow mesenchymal stem cell’s differentiation into neural-like cell induced by Valproic Acid. VPA enhanced the Oct-4 in vitro directed differentiation.
