Fungi have been regarded as an attractive source for discovering new bioactive compounds nowadays. Some bioactive metabolites from fungi, such as cephalosporins, penicillins, cephalosporins, mevastatin and lovastatin, have been the crucial products of bio-pharmaceutical industry. For the sake of looking for bioactive metabolites from fungi, we put fungus Stachybotrys parvispora strain HS-FG-843 as the experiment target to undertake cultivation of the stain, fermentation, isolation and purification, structure elucidation and the cytotoxicity in order to find more bioactive metabolites. In this paper, we first conducted an fermentation experiment of soil fungi with 250 ml erlenmeyer flasks. Then, activity detection test was carried out on the crude extract of broth by combined antitumor detection model of human lung cell line A549 in vitro with TLC detection model. As a consequence, we selected a Stachybotrys parvispora strain HS-FG-843 exhibited better activity. Therefore, the fermentation of HS-FG-843 was amplifid by 30 L fermenter. Finally, 10 compounds were obtained by the method of isolation and purification including the pre-treatment of separate the fermentation broth, Diaion HP20 resin column, silica gel column, Sephadex LH-20 gel column and semi- preparative HPLC. There are a new compound among them. We also elucidated their stuctures on the basis of spectroscopic methods(1H-NMR, 13C-NMR). Give name to the new compound respectively : stachybotrin G. we detected the cytotoxicity of all the obtained compounds against A549 cell lines in vito through the CCK8 method. The result indicated that two new compounds had better cytotoxicity through the inhibition ratio. As a result, stachybotrin G exhibited cytotoxic activity with IC50 values of 17.4μg/ml.
